摘  要:目的探讨益肾丸治疗慢性肾小球肾炎(CGN)的活性成分和药效机制。方法通过TCMSP检索益肾丸的潜在活性成分和靶点,通过GeneCards数据库获取CGN的靶点,将共同靶点与潜在活性成分导入Cytoscape 3.7.2软件构建中药成分共同靶点疾病相互作用网络,并获得核心成分,通过STRING 11.0数据库获得共同靶点蛋白质蛋白质相互作用网络及核心靶点;通过Metascape数据库进行GO功能富集和KEGG通路富集分析。运用AutoDock Vina软件对核心成分及核心靶点进行分子对接验证。结果筛选获得益肾丸115个潜在活性成分,CGN与2765个靶点相关,潜在活性成分与疾病有135个共同靶点。核心成分包括槲皮素、木犀草素等,核心靶点包括TNF、IL6、AKT1等。KEGG富集分析主要涉及癌症的发病途径等。分子对接结果显示,核心成分与核心靶点结合具有较好的活性。结论益肾丸治疗CGN的主要机制可能通过槲皮素等核心成分作用于TNF等靶点发挥作用,为进一步阐明益肾丸的药效物质基础及作用机制奠定了基础。Objective To explore the active ingredients and pharmacodynamic mechanism of Yishen pill in treating chronic glomerulonephritis(CGN).Methods The potential active ingredients and related targets of Yishen pill were searched through TCMSP.The targets of CGN were obtained from GeneCards.The common targets and potential active ingredients were imported into Cytoscape 3.7.2 software to construct the interaction network of herbs ingredients common targets disease and obtain the core active ingredients.The protein-protein interaction network of common targets and core targets were obtained by STRING 11.0 database.GO functional enrichment and KEGG pathway enrichment analyses were performed using Metascape database.Core active ingredients and core targets were selected for molecular docking verification by AutoDock Vina software.Results A total of 115 potential active ingredients of Yishen pill and 2765 targets of CGN were screened,including 135 common targets between potential active ingredients and disease.Core ingredients included quercetin,luteolin,etc.Core targets included TNF,IL6,AKT1,etc.KEGG enrichment analysis mainly involved pathways in cancer,etc.The results of molecular docking showed that the core ingredients had good binding activity with the core targets.Conclusion Yishen pill may act on TNF and other targets through quercetin and other core ingredients in the treatment of CGN,which lays a foundation for further elucidating the pharmacodynamic material basis and mechanism of Yishen pill.
