摘  要:肿瘤免疫治疗当前已成为肿瘤临床治疗的一大利器。然而,免疫逃逸是制约肿瘤免疫治疗疗效的瓶颈问题,导致肿瘤免疫逃逸的关键环节之一是CD8^(+)T细胞耗竭。研究表明,CD8^(+)T细胞在肿瘤微环境各种免疫抑制性因素的作用下发生表观重编程,导致CD8^(+)T细胞耗竭。如何有效靶向调节表观重编程机制抑制CD8^(+)T细胞耗竭,是目前亟待解决的重点和难点问题。本文综述了肿瘤微环境CD8^(+)T细胞耗竭的特征以及在肿瘤免疫逃逸中的表观重编程机制,并阐明了靶向调节表观遗传机制改善CD8^(+)T细胞耗竭的策略,旨在为肿瘤免疫治疗提供新思路、开辟新路径。At present,tumor immunotherapy has become a powerful weapon in clinical tumor treatment.However,immune escape is a bottleneck that restricts the efficacy of tumor immunotherapy,and CD8^(+)T cell exhaustion is one of the crucial causes in tumor immune escape.Researches have shown that CD8^(+)T cells undergo epigenetic reprogramming under the influence of various immunosuppressive factors in the tumor microenvironment,which leads to CD8^(+)T cell exhaustion.How to effectively target and regulate epigenetic reprogramming mechanisms to inhibit CD8^(+)T cell exhaustion is currently a key and difficult problem that needs to be solved urgently.This paper reviews the characteristics of CD8^(+)T cell exhaustion in the tumor microenvironment and epigenetic reprogramming mechanisms in tumor immune escape.It also elucidates the strategies for improving CD8^(+)T cell exhaustion through targeted regulation of epigenetic mechanisms,and aims to provide new ideas and open up new pathways for tumor immunotherapy.
