摘 要:目的:探究精神分裂症长链非编码RNA-微小RNA-信使RNA(long non-coding RNA-microRNA-messenger RNA,lncRNA-miRNA-mRNA)调控网络,明确疾病发病机制。方法:采用高通量测序技术检测精神分裂症患者和健康人群血液中RNA表达水平,以校正统计值q<0.05为阈值筛选差异RNA,并构建lncRNA-miRNA-mRNA调控网络;对网络中mRNA进行富集分析、蛋白互作分析并筛选核心基因。结果:共筛选出157个lncRNA,43个miRNA,2133个mRNA在精神分裂症中的差异有统计学意义,其中54个lncRNA,11个miRNA,153个mRNA构建了竞争性内源RNA调控网络。网络中的mRNA富集于NF-κB信号通路、泛素介导的蛋白水解等信号通路;TBL3、FTSJ3、BCL2L1、UBE2I和CUL4A共5个核心靶基因在网络中占据重要位置。结论:调控网络构建有助于进一步认识精神分裂症分子机制,筛选潜在的生物标志物,为疾病后续的干预治疗提供新的作用靶点。Objective:To explore the regulatory network of long non-coding RNA-microRNA-messenger RNA(lncRNA-miRNA-mRNA)in schizophrenia and elucidate the pathogenesis.Method:High-throughput sequencing technology was used to measure RNA expression levels in the blood of schizophrenia patients and healthy populations.Differential expression analysis was conducted using a q-value threshold of<0.05,and lncRNA-miRNA-mRNA regulatory network was constructed.The enrichment analysis and protein-protein interaction analysis of mRNA in the network was performed,and the key genes were identified.Results:A total of 157 lncRNAs,43 miRNAs,and 2133 mRNAs were statistically significant in schizophrenia.Among them,a ceRNA regulatory network consisting of 54 lncRNAs,11 miRNAs,and 153 mRNAs was constructed.The mRNA in the network was enriched in signaling pathways such as the NF-κB pathway,ubiquitin-mediated protein degradation.TBL3,FTSJ3,BCL2L1,UBE2I and CUL4A,which are 5 core target genes,played crucial roles in the network.Conclusion:The construction of the regulatory network contributes to a better understanding of the molecular mechanisms of schizophrenia,facilitates the identification of potential biomarkers,and provides new targets for intervention and drug screening in subsequent disease treatment.
