青少年抑郁症非自杀性自伤行为与色氨酸羟化酶2基因多态位点及负性生活事件交互作用的关联

摘　　要：目的:探讨色氨酸羟化酶2基因位点rs11178997、rs11178998、rs120074175多态性及负性生活事件交互作用与青少年抑郁症非自杀性自伤行为的关联。方法:提取江苏淮安地区197例青少年抑郁症患者(非自杀性自伤组106例、无非自杀性自伤组91例)基因组DNA,采用聚合酶链式反应技术扩增DNA片段,检测两组患者的基因型。应用青少年生活事件量表对两组进行问卷调查。采用χ2检验分析两组等位基因以及基因型的频率分布差异,广义多因素降维法(generalized multifactor dimensionality reduction,GMDR)分析基因和环境的交互作用。结果:非自杀性自伤组与无非自杀性自伤组在人际关系因子、受惩罚因子差异均有统计学意义(P均<0.05)。GMDR分析发现rs11178997、rs11178998和青少年生活事件之间存在相互作用。结论:青少年抑郁症患者尤其女性面临更多的受惩罚和人际关系困扰时可能会倾向采用自伤行为来应对情绪,rs11178997、rs11178998多态位点与生活事件交互作用可能与非自杀性自伤发生存在联系。Objective:To explore the association between tryptophan hydroxylase 2 single nucleotide polymorphism(rs11178997,rs11178998,rs120074175)and negative life events interactions and non-suicidal self-injury(NSSI)in adolescents with depression.Method:Genomic DNA was extracted from 197 adolescents with depression in Huai'an(106 in the NSSI group and 91 in the non-NSSI group).DNA fragments were amplified by polymerase chain reaction technology to detect the genotypes of the NSSI group and the non-NSSI group.Questionnaire survey was conducted on the two groups using the adolescent life events scale.The frequency distribution differences of alleles and genotypes of the NSSI group and the non-NSSI group were analyzed byχ2 test,and the interaction between gene and environment was analyzed by generalized multifactor dimensionality reduction(GMDR).Results:There were statistically significant differences in interpersonal relationship factor and punishment factor between the NSSI group and the non-NSSI group(all P<0.05).GMDR analysis found that there was an interaction between rs11178997,rs11178998 and adolescent life events.Conclusion:Adolescents with depression,especially females,may tend to adopt NSSI behavior to cope with emotions when facing more family and interpersonal relationship problems.The interaction between rs11178997,rs11178998 polymorphisms and life events may be associated with the occurrence of NSSI.
