摘  要:为提高沙棘叶副产物利用率,研发并优化适于工业化生产的沙棘叶总黄酮提取工艺。以总黄酮得率为响应值,在单因素试验的基础上,采用响应面试验优化亚临界水提取沙棘叶总黄酮工艺,并与溶剂提取法和超声提取法进行比较。结果表明,4个单因素中,提取温度、料液比、提取时间对沙棘叶总黄酮得率影响较大,依次分别为:提取温度>料液比>提取时间,但提取压力对其无显著影响;响应面优化得到最优工艺参数为:提取温度130℃,提取时间19 min,料液比1∶26(g/mL),在此条件下,沙棘叶总黄酮得率可达6.87%±0.15%,分别为传统溶剂提取法和超声提取法的1.95倍和1.81倍。本研究为生产中沙棘叶总黄酮的提取提供了一种绿色高效的方法,也为其开发利用提供了新思路。In order to improve the utilization rate of seabuckthorn leaves(SBL)by-products,the extraction process of total flavonoids from SBL suitable for industrial production was developed and optimized.Using the yield of total flavonoids as the response value,a response surface test was used to optimize the subcritical water extraction(SWE)process of total flavonoids from SBL based on single-factor experiments,and comparisons were made with solvent extraction and ultrasonic extraction methods.The results showed that among the four single factors,extraction temperature,material-to-liquid ratio and extraction time had a great influence on the yield of total flavonoids from SBL,in the order of:extraction temperature>material-to-liquid ratio>extraction time,but extraction pressure had no significant effect on it.The optimum parameters obtained from the response surface optimization were:an extraction temperature of 130℃,an extraction time of 19 min,and a material-to-liquid ratio of 1∶26(g/mL).Under these conditions,the yield of total flavonoids from SBL reached 6.87%±0.15%,which was 1.95 times and 1.81 times that of the traditional solvent extraction method and ultrasonic extraction method,respectively.The results provide a green and efficient extraction pathway for the extraction of total flavonoids from SBL in production and also afford a new idea for its development and utilization.
