摘 要:目的探讨序列相似家族83成员A(FAM83A)对乳腺癌细胞增殖和迁移的作用及其可能机制。方法将人乳腺癌细胞株MCF-7分为FAM83A过表达组(O-FAM83A组)、FAM83A沉默组(sh-FAM83A组)、空白对照组(Blank组)、过表达的阴性对照组(O-NC组)及沉默的阴性对照组(sh-NC组)。采用实时荧光定量PCR检测FAM83A mRNA表达,CCK-8法和划痕实验分别检测MCF-7细胞增殖和迁移,Western blot法检测转化生长因子-β1(TGF-β1)、磷酸化Smad3(p-Smad3)、细胞周期蛋白D1(Cyclin D1)、MMP-2和MMP-9蛋白表达。结果与Blank组和O-NC组相比,O-FAM83A组FAM83A mRNA表达升高(P<0.01),不同时间点细胞增殖和细胞划痕愈合率差异无统计学意义(P>0.05)。与Blank组和sh-NC组相比,sh-FAM83A组FAM83A mRNA表达降低(P<0.01),不同时间点细胞增殖差异无统计学意义(P>0.05),48和72h时细胞划痕愈合率升高(P<0.05),TGF-β1、p-Smad3、Cyclin D1、MMP-2和MMP-9蛋白表达均降低(P<0.05)。结论沉默FAM83A可能通过抑制TGF-β1/Smad3信号通路来促进乳腺癌细胞的迁移。Objective To investigate the effect and underlying mechanism of family with sequence similarity 83 member A(FAM83A)on the proliferation and migration of breast cancer cells.Methods Human breast cancer cell line MCF-7 was divided into O-FAM83A group,sh-FAM83A group,Blank group,O-NC group and sh-NC group.The mRNA expression of FAM83A was determined by qRT-PCR.The proliferation and migration of MCF-7 cells were measured by CCK-8 and cell scratch assays,respectively.The protein expressions of TGF-β1,phosphorylated Smad3(p-Smad3),Cyclin D1,MMP-2 and MMP-9 were detected by Western blot.Results Compared with Blank group and O-NC group,the mRNA expression of FAM83A was increased(P<0.01),while the cell proliferation and healing rate of cell scratch at different time points were not significantly different in O-FAM83A group(P>0.05).Compared with Blank group and sh-NC group,the mRNA expression of FAM83A was decreased(P<0.01),the cell proliferation was not obviously changed(P>0.05),the healing rates of cell scratch after 48 and 72 hours were increased(P<0.05),and the protein expressions of TGF-β1,p-Smad3,Cyclin D1,MMP-2 and MMP-9 were downregulated in sh-FAM83A group(P<0.05).Conclusion Silencing FAM83A may promote the migration of breast cancer cells by inhibiting TGF-β1/Smad3 signaling pathway.
