摘 要:目的:观察阿魏酸和绿原酸对特应性皮炎的体表优势菌群(金黄色葡萄球菌、表皮白葡萄球菌)以及相关细胞(HaCaT细胞、RAW264.7细胞)活性的影响。方法:采用微量液体倍比稀释法配制阿魏酸和绿原酸药液,浓度为60、120、240、480、960、1920、3840、7680、15360μmol·L^(-1)。取金黄色葡萄球菌和表皮白葡萄球菌,加入阿魏酸或绿原酸药液进行干预。采用MMT法检测细菌活性,结晶紫染色法检测细菌生物膜活性。取HaCaT细胞和RAW264.7细胞(0.1%脂多糖诱导),加入阿魏酸或绿原酸药液进行干预,并采用MTT法检测细菌活性。结果:MTT检测显示,阿魏酸浓度为15360μmol·L^(-1)时,金黄色葡萄球菌和表皮白葡萄球菌活性降低(P<0.05)。阿魏酸浓度为960μmol·L^(-1)和1920μmol·L^(-1)时,金黄色葡萄球菌活性增强(P<0.05)。绿原酸浓度为15360μmol·L^(-1)时,表皮白葡萄球菌活性降低(P<0.00005)。阿魏酸和绿原酸浓度为3840、7680、15360μmol·L^(-1)时,金黄色葡萄球菌生物膜活性降低(P<0.05)。阿魏酸浓度为960μmol·L^(-1)、15360μmol·L^(-1)时,表皮白葡萄球菌生物膜活性增强(P<0.05)。绿原酸浓度为60μmol·L^(-1)、15360μmol·L^(-1)时,表皮白葡萄球菌生物膜活性降低(P<0.05)。绿原酸浓度为1920μmol·L^(-1)时,HaCaT细胞存活率降低(P<0.0005)。绿原酸浓度为30、120、240、480、960、1920μmol·L^(-1)时,RAW264.7细胞存活率降低(P<0.005)。结论:阿魏酸和绿原酸对特应性皮炎的2种优势菌群(金黄色葡萄球菌和表皮白葡萄球菌)具有一定的抑制作用。Objective:To observe the effects of ferulic acid and chlorogenic acid on the activities of dominant flora(Staphylococcus aureus and Staphylococcus epidermidis)and related cells(HaCaT cells and RAW264.7 cells)in atopic dermatitis(AD).Methods:Ferulic acid and chlorogenic acid liquid were prepared by microliquid dilution method with concentrations of 60,120,240,480,960,1920,3840,7680,15360μmol·L^(-1).Staphylococcus aureus and Staphylococcus albicans epidermis were added to the solution of ferulic acid or chlorogenic acid for intervention.The MMT method was used to detect bacterial activity,and the crystal violet staining method was used to detect bacterial biofilm activity.HaCaT cells and RAW264.7 cells(0.1%lipopolysaccharide induction)were added to the ferulic acid or chlorogenic acid solution for intervention,and the bacterial activity was detected by MTT method.Results:MTT showed that the concentration of ferulic acid was 15360μmol·L^(-1),the activities of Staphylococcus aureus and Staphylococcus albicans epidermis decreased(P<0.05).The concentration of ferulic acid was 960μmol·L^(-1)and 1920μmol·L^(-1),the activity of Staphylococcus aureus was enhanced(P<0.05).The concentration of chlorogenic acid was 15360μmol·L^(-1),the activity of Staphylococcus albicans epidermis decreased(P<0.00005).The concentrations of ferulic acid and chlorogenic acid were 3840,7680 and 15360μmol·L^(-1),the biofilm activity of Staphylococcus aureus decreased(P<0.05).The concentration of ferulic acid was 960μmol·L^(-1),15360μmol·L^(-1),the biofilm activity of Staphylococcus albicans epidermis was enhanced(P<0.05).The concentration of chlorogenic acid was 60μmol·L^(-1),15360μmol·L^(-1),the biofilm activity of Staphylococcus albicans epidermis decreased(P<0.05).The concentration of chlorogenic acid was 1920μmol·L^(-1),the viability of HaCaT cells decreased(P<0.0005).The concentrations of chlorogenic acid were 30,120,240,480,960,1920μmol·L^(-1),the survival rate of RAW264.7 cells decreased(P<0.005).Conclusion:Ferulic
