摘　　要：目的探讨CDCA8在肺腺癌中的表达及其免疫调控机制。方法UALCAN数据库下载肺腺癌和正常肺组织的转录组学数据,比较肺腺癌和正常肺组织中CDCA8 mRNA表达水平的差异。UALCAN数据库在线筛选肺腺癌中与CDCA8的共表达基因,并对共表达基因在DAVID数据库中进行GO和KEGG富集分析。K-M数据库在线分析肺腺癌组织中CDCA8 mRNA表达水平与总体生存率的关系。TIMER数据库分析肺腺癌中CDCA8 mRNA表达水平与多个免疫细胞浸润程度的相关性。HPA数据库验证CDCA8蛋白表达水平在肺腺癌组织及正常肺组织中的差异。纳入2023年1月-2024年1月本院肿瘤科的78例肺腺癌患者(肺腺癌组)以及50名健康者(对照组)作为研究对象,ELISA法检测并比较两组血清中CDCA8蛋白表达水平的差异。血清中CDCA8表达水平在鉴别肺腺癌中的临床效能采用ROC曲线分析。结果肺腺癌组织中CDCA8 mRNA中位表达水平为10.80(5.76,18.09),显著高于正常肺组织CDCA8 mRNA表达水平[1.36(0.97,1.72)],差异具有统计学意义(P<0.001)。肺腺癌CDCA8 mRNA高表达组和低表达组中位生存时间分别为59个月和106个月,肺腺癌CDCA8 mRNA高表达组生存率显著低于低表达组,差异具有统计学意义(HR=1.66,P<0.001)。肺腺癌中与CDCA8具有显著正相关性的基因共200个,与CDCA8具有显著负相关性的基因共107个,307个差异共表达基因的GO和KEGG富集分析于细胞分裂、G2/M有丝分裂细胞周期转变、细胞核、细胞、微管结合、蛋白结合、DNA结合、细胞周期、DNA复制等。相关性分析结果显示,肺腺癌中CDCA8 mRNA表达水平与CD8+T细胞、CD4+T细胞、巨噬细胞、树突状细胞浸润程度无明显相关性(P>0.05),与B细胞浸润程度具有显著负相关性(r=-0.158,P<0.001),与中性粒细胞浸润程度具有显著正相关性(r=0.131,P<0.001)。免疫组化结果显示,HPA 028120、HPA 028258、HPA 028783三种抗体染色后肺腺癌组织中CDCA8蛋白低度和Objective To investigate the expression level of CDCA8 in lung adenocarcinoma and its immune regulation mechanisms.Methods The transcriptomic data of lung adenocarcinoma and normal lung tissues were downloaded from UALCAN database,and the difference of CDCA8 mRNA expression between lung adenocarcinoma and normal lung tissues were compared.The co-expressed genes of CDCA8 in lung adenocarcinoma were screened online by UALCAN database,and the co-expressed genes were further enriched by GO and KEGG analysis in DAVID database.The K-M database was used to analyze the relationship between CDCA8 mRNA expression level and overall survival rate in lung adenocarcinoma patients.The correlation between the expression level of CDCA8 mRNA and the infiltration degree of multiple immune cells were analyzed by TIMER database.The HPA database was used to verify the difference of CDCA8 protein expression level in lung adenocarcinoma tissue and normal lung tissues.A total of 78 patients with lung adenocarcinoma(lung adenocarcinoma group)and 50 healthy controls(control group)from the department of oncology,the Nanyang Central Hospital between January 2023 and January 2024 were enrolled in the study.The ELISA method was used to detect and compare the difference of CDCA8 protein expression level between the lung adenocarcinoma group and control group.The clinical efficacy of serum CDCA8 expression level in differentiating lung adenocarcinoma from the healthy controls was analyzed by ROC curve.Results The median expression level of CDCA8 mRNA in lung adenocarcinoma tissues was 10.80(5.76,18.09),which was significantly higher than 1.36(0.97,1.72)of normal lung tissues,the difference was statistically significant(P<0.001).The median survival time of the CDCA8 mRNA high and low expression group was 59 months and 106 months,respectively.The survival rate of the CDCA8 mRNA high expression group was significantly lower than that of the CDCA8 mRNA low expression group,with statistical significance(HR=1.66,P<0.001).There were 200 genes positivel
