摘  要:研究不同产地(S1~S7)“宁杞1号”枸杞类胡萝卜素HPLC指纹图谱与其抗氧化活性的谱-效关系。采用《中药指纹图谱相似度评价系统(2012年版)》建立不同产地枸杞类胡萝卜素指纹图谱,并进行共有峰指认、相似度评价和聚类分析。以DPPH、羟基自由基清除率为抗氧化指标,采用灰色关联度法分析枸杞类胡萝卜素与抗氧化指标的谱-效关系。结果显示:枸杞类胡萝卜素共10个共有峰,指认了1、2、4、10号峰分别为玉米黄素、β-隐黄质、β-类胡萝卜素、玉米黄素双棕榈酸酯,且7个产地枸杞类胡萝卜素组成成分相似,含量最高的均为玉米黄素双棕榈酸酯。不同产地枸杞抗氧化活性差异较大,宁夏中宁(S1)、宁夏兴仁(S2)、青海格尔木(S4)所产的“宁杞1号”枸杞类胡萝卜素抗氧化能力显著强于其他4个产地。谱-效关系分析表明:除1号峰外,其余共有峰与DPPH自由基清除率的关联度均大于0.6,且10号峰与DPPH自由基清除率的关联度大于0.8;10个共有峰与羟基自由基清除率的关联度在0.60~0.72之间,且10号峰与羟基自由基清除率的关联度最大,可达0.72。由此说明,枸杞类胡萝卜素发挥抗氧化作用的基础可能是其各共有峰所代表的化学成分共同作用的结果,其中10号峰所代表的组分——玉米黄素双棕榈酸酯可能是枸杞类胡萝卜素抗氧化作用的主要活性成分。HPLC fingerprints of carotenoids of"Ningqi No.1"Lycium barbarum from different origin(S1-S7)were studied,and the relationship between their spectral activity and antioxidant activity was studied.The carotenoid fingerprints of Lycium barbarum from different regions were established by the Similarity Evaluation System of TCM Fingerprint(2012 edition),and the common peak identification,similarity evaluation and cluster analysis were carried out.Using DPPH and hydroxyl radical clearance as antioxidant indexes,the spectrum-effect relationship between carotenoids and antioxidant indexes of Lycium barbarum was analyzed by grey correlation degree method.The results showed that there were 10 common peaks of carotenoids in Lycium barbarum,and the peaks 1,2,4 and 10 were identified as zeaxanthin,β-cryptoxanthin,β-carotenoid and zeaxanthin dipalmitate,respectively.The composition of carotenoids was similar in the 7 producing areas,and the highest content was zeaxanthin dipalmitate.The antioxidant activity of Lycium barbarum from different producing areas was significantly different.The antioxidant capacity of carotenoid of"Ningqi No.1"from Zhongning,Ningxia(S1),Xingren,Ningxia(S2)and Golmu,Qinghai(S4)was significantly higher than that from the other four producing areas.The spectrum-effect relationship analysis indicated that,except for peak 1,the correlation coefficients between the other common peaks and the DPPH radical clearance rate were all greater than 0.6,and the correlation coefficient between peak 10 and the DPPH radical clearancerate was greater than 0.8.The correlation coefficients between the 10 common peaks and the hydroxyl radical clearance rate ranged from 0.60 to 0.72,and the correlation coefficient between peak 10 and the hydroxyl radical clearance rate was the highest,reaching 0.72.This suggests that the basis for the antioxidant effect of Lycium barbarum carotenoids may be the combined action of the chemical components represented by each common peak,among which the component represented by peak 10-zeax
