摘 要:琥珀酸盐作为线粒体三羧酸循环的中间代谢物,其代谢作用被广泛研究。近年来,越来越多的研究集中在琥珀酸盐积累导致相关肿瘤发生的分子机制上,例如作为炎症信号和诱导表观遗传改变。实际上,琥珀酸脱氢酶基因突变和异常琥珀酸盐积累已经在一系列遗传性和散发性的恶性肿瘤中被观察到。该文阐述了肿瘤微环境中高浓度的琥珀酸盐在肿瘤发生进程中所发挥的作用,并深入探讨了琥珀酸盐的作用和机制。以机制研究为重点,总结了琥珀酸盐作为癌症的诊断标志物和已经应用于临床或正在开发的靶向治疗方法。由此推测,针对不同分子靶点研发的药物,有望拓展抗癌治疗领域的研究视野。The metabolic role of succinate as an intermediate metabolite in the mitochondrial tricarbox-ylic acid cycle has been extensively studied.In recent years,an increasing number of studies have focused on the molecular mechanisms by which succinate accumulation leads to associated tumourigenesis,as an inflammatory signal and induction of epigenetic alterations.Indeed,mutations in the succinate dehydrogenase gene and aberrant succinate accumulation have been observed in a range of hereditary and sporadic malignancies.The paper describes the role played by high concentrations of succinate in the tumour microenvironment in the process of tumourigen-esis and provide insights into the role and mechanisms of succinate.With a focus on mechanistic studies,the article summarises the use of succinate as a diagnostic marker for cancer and targeted therapies that have been applied in the clinic or are under development.It is surmised that drugs developed for different molecular targets are expected to expand the research horizons in the field of anti-cancer therapy.
