摘 要:目的:探讨黄芩苷对根尖周炎大鼠炎症和骨吸收的影响,并基于音猬因子(SHH)信号通路分析其作用机制。方法:将50只SD大鼠随机分为对照组、模型组、0.01%黄芩苷组、0.1%黄芩苷组、1%黄芩苷组,每组10只。除对照组外,其余各组采用髓腔直接暴露法建立根尖周炎大鼠模型。Micro-CT扫描评估牙槽骨吸收情况;HE染色观察大鼠根尖周组织病理学改变;抗酒石酸酸性磷酸酶(TRAP)染色检测大鼠根尖周组织破骨细胞数量;ELISA法检测大鼠根尖周组织炎症因子[肿瘤坏死因子-α(TNF-α)、白细胞介素(IL)-6、IL-1β]水平;RT-qPCR检测大鼠根尖周组织骨代谢相关基因[骨保护素(OPG)、核因子κB受体活化因子配体(RANKL)、基质金属蛋白酶-9(MMP-9)]表达;Western blot检测大鼠根尖周组织SHH信号通路相关蛋白(SHH、Ptch1、Smo、Gli1)表达水平。结果:与对照组比较,模型组大鼠骨小梁分离度(Tb.Sp)、根尖周病灶体积、破骨细胞数量、炎症因子水平、SHH信号通路相关蛋白表达水平、RNAKL、MMP-9 mRNA表达水平及RNAKL/OPG比值升高,骨小梁厚度(Tb.Th)及OPG mRNA表达水平降低(均P<0.05),HE染色显示根尖周组织炎症细胞浸润增多。与模型组比较,0.1%、1%黄芩苷组上述指标明显改善(均P<0.05),HE染色显示根尖周组织炎症细胞浸润减少。结论:黄芩苷可能通过抑制SHH信号通路,减少破骨细胞生成,减轻牙周炎症及牙槽骨吸收,从而改善大鼠根尖周炎。Objective:To investigate the effect of baicalin on inflammation and bone resorption in rats with periapical periodontitis and to elucidate its mechanism of action based on the Sonic hedgehog(SHH)signaling pathway.Methods:Fifty SD rats were randomly divided into five groups:control group,model group,0.01%baicalin group,0.1%baicalin group and 1%baicalin group,with 10 rats in each group.Except for the control group,the other groups were used to establish periapical periodontitis rat model through direct pulp exposure.Micro-CT scanning was used to assess alveolar bone resorption,and HE staining was performed to observe histopathological changes in the periapical tissues.Tartrate-resistant acid phosphatase(TRAP)staining was used to detect the number of osteoclasts in the periapical tissues.The levels of inflammatory cytokines(tumor necrosis factor-α[TNF-α],interleukin[IL]-6,IL-1β)in rat periapical tissues were detected by ELISA.RT-qPCR was employed to detect the expression of bone metabolism-related genes(osteoprotegerin[OPG],receptor activator of nuclear factor-κB ligand[RANKL],matrix metallopeptidase-9[MMP-9]),and Western blot was used to analyze the expression of SHH signaling pathway-related proteins(SHH,Ptch1,Smo,Gli1).Results:Compared with the control group,the model group exhibited increased Tb.Sp,periapical lesion volume,number of osteoclasts,levels of inflammatory cytokines,expression levels of SHH signaling pathway-related proteins,RANKL and MMP-9 mRNA expression levels,and RANKL/OPG ratio,while Tb.Th and OPG mRNA expression levels were decreased(all P<0.05).Histopathological examination by HE staining revealed increased inflammatory cell infiltration in the periapical tissues of the model group.Compared with the model group,the 0.1%and 1%baicalin groups showed significant improvements in these parameters(all P<0.05),with reduced inflammatory cell infiltration in the periapical tissues as indicated by HE staining.Conclusion:Baicalin may reduce osteoclastogenesis and alleviate inflammation and alveolar bo
