癫痫发作期神经胶质细胞炎性反应情况及与Wnt3a信号分子表达的关系

摘  要:目的探究癫痫发作期神经胶质细胞的炎性反应及与髓系细胞触发受体2(TREM2)和Wnt3a表达的关系。方法利用腹腔注射氯化锂-匹鲁卡品成功制备SD大鼠癫痫模型(Se1d、Se2d、Se3d、Se7d和Se14组)和建立体外Astrocytes/Microglia细胞共培养体系。应用免疫荧光染色测定TREM2和Wnt3a的阳性表达情况。应用Western blot蛋白分析法测定TREM2和Wnt3a的表达量变化情况。结果蛋白电泳结果显示,在癫痫发作的较早期,与Con组(注射相同剂量的生理盐水)相比,GFAP(Se1d、Se2d和Se3d组)和IBA1(Se1d和Se2d组)蛋白表达量上调(P均<0.05);免疫荧光结果显示,TREM2只在小胶质细胞的细胞膜上表达,Wnt3a在星形胶质细胞和小胶质细胞的细胞核上均有表达,且癫痫发作后TREM2和Wnt3a阳性细胞数明显增加;蛋白电泳结果显示,与Con组相比,癫痫各组TREM2的蛋白表达量上调(P均<0.01);与Con组相比,癫痫各组Wnt3a的蛋白表达量增加(P<0.001)。结论SD大鼠癫痫发作后诱发神经炎性反应,神经胶质细胞活化,小胶质细胞TREM2被二次激活,Wnt3a信号可能参与癫痫神经炎性的调控。Objective To explore the inflammatory response of glial cells during epileptic seizures and its relationship with TREM2 and Wnt3a expression.Methods The epileptic rat model was successfully induced by intraperitoneal injection of lithium chloride-pilocarpine(Se1d,Se2d,Se3d,Se7d and Se14d groups),and astrocytes and microglia were co-cultured in vitro.The positive expressions of TREM2 and Wnt3a were detected by immunofluorescence staining.The changes of TREM2 and Wnt3a protein levels were measured by Western blot.Results The Western blot results showed that the protein contents of GFAP of Se1d group,Se2d group and Se3d group and IBA1 of Se1d group and Se2d group were significantly up-regulated in the early stage of epileptic seizure(within 3 d),and the difference was statistically significant(P all<0.05).The immunofluorescence results indicated that TREM2 was only expressed on the membrane of microglia,and Wnt3a was expressed on the nucleus of astroglia and microglia,and the number of TREM2 and Wnt3a positive cells increased significantly after epileptic seizure.The protein results showed that the expression of TREM2 protein in epileptic groups was significantly upregulated(P all<0.01).Meanwhile the Wnt3a protein expression in epileptic group was also significantly increased(P<0.001).Conclusion After epileptic seizures in SD rats,neuroinflammatory reactions are induced,with activation of glial cells and secondary activation of TREM2 in microglia.The Wnt3a signaling may be involved in the regulation of neuroinflammatory epilepsy.
