摘 要:十溴联苯醚(BDE-209)因其优异的阻燃性能和低廉的成本,被广泛应用于商业领域,并在环境中普遍检出。尽管近年来BDE-209对水生生物的毒性研究日益增多,但其对鱼类脂质代谢的影响研究匮乏,相关分子机制尚不明确。因此,本研究开展了不同浓度BDE-209(0、1、10、100μg·L^(-1))的斑马鱼胚胎急性毒性测试,利用油红O染色表征了不同发育阶段斑马鱼体内中性脂质的积累变化,明确了BDE-209对斑马鱼胚胎/仔鱼脂质代谢干扰的效应时间窗口;进一步通过甘油三酯(triglyceride,TG)和总胆固醇(total cholesterol,TC)含量测定、脂质组学分析及基因表达验证,揭示了BDE-209暴露对斑马鱼胚胎/仔鱼脂质代谢影响的潜在机制。研究结果表明,100μg·L^(-1)BDE-209暴露显著降低了斑马鱼存活率和体长,加速了仔鱼心跳并抑制其卵黄囊的吸收利用;此外,斑马鱼体内TG和TC等中性脂质浓度显著上升;脂质组学分析显示,BDE-209暴露可引起包括鞘脂类、甘油磷脂类在内的74种脂质分子的显著积累;基因表达结果表明,BDE-209暴露显著上调了脂质合成相关基因CDP-甘油二酯-肌醇-3-磷脂酰转移酶(cdipt)溶血心磷脂酰基转移酶1(lclat1)和磷脂酸膦酸水解酶(lpin1a)的表达,显著下调了磷脂酶D1(pld1)、磷脂酶A2(pla2g12a)基因的表达。综上所述,BDE-209通过影响斑马鱼早期发育过程中脂质代谢相关基因的表达,干扰脂质代谢过程,造成脂质积累,从而引发斑马鱼早期生命阶段的发育毒性。Decabromodiphenyl ether(BDE-209)is widely used in commercial applications due to its excellent flame retardant properties and low cost.Although researches on the toxicity of BDE-209 to aquatic organisms have increased in recent years,studies on its effects on lipid metabolism and early development in fish remain limited.Furthermore,the underlying molecular mechanisms are still unclear.Therefore,we conducted an acute toxicity t esting using zebrafish embryos exposed to different concentrations of BDE-209(0,1,10 and 100μg·L^(-1)).We used Oil Red O staining to characterize the changes of neutral lipid accumulation in zebrafish embryos/larvae at different developmental stages,and identified the temporal window of lipid metabolism disruption caused by B DE-209.Furthermore,triglyceride(TG)and total cholesterol(TC)contents,lipidomics profiling,and gene e xpression validation were conducted to elucidate the potential mechanisms of BDE-209 in the disruption of lipid metabolism in zebrafish embryos/larvae.The results demonstrated that exposure to 100μg·L^(-1)BDE-209 s ignificantly reduced the survival rate and body length of zebrafish larvae,accelerated heart rate in larvae,and i nhibited the absorption and utilization of yolk sac.Additionally,the concentrations of neutral lipids,such as TG and TC,were significantly increased.Lipidomics analysis revealed that BDE-209 exposure led to the significant a ccumulation of 74 lipid molecules,such as sphingolipids and glycerophospholipids.Gene expression results i ndicated that BDE-209 exposure significantly up-regulated the expressions of lipid synthesis-related genes,i ncluding CDP-diacylglycerol-inositol 3-phosphatidyltransferase(cdipt),lysocardiolipin acyltransferase 1(lclat1)a nd phosphatidic acid phosphohydrolase(lpin1a),and significantly down-regulated the expressions of phospholipase D1(pld1)and phospholipase A2(pla2g12a).In conclusion,BDE-209 disrupted the process of lipid metabolism in z ebrafish embryos/larvae by affecting the expressions of lipid metabolism-related
