摘 要:通过连续14 d的小鼠摄食贝类暴露实验以获得砷在生物体中的累积、代谢、转化情况。结果表明膳食暴露14 d后,小鼠摄入的总砷最终约70%以粪便形式排出。总砷在各脏器/组织中的蓄积为胃(0.26~0.54μg·g^(-1))>肠(0.12~0.42μg·g^(-1))>脾(0.031~0.162μg·g^(-1))>肾(0.06~0.12μg·g^(-1))>肝脏(0.049~0.091μg·g^(-1)),在其余脏器/组织中的蓄积不明显(0.016~0.065μg·g^(-1));食物中的砷甜菜碱(AsB)在暴露14 d后约有46%以粪便形式排出,而二甲基砷(DMA)约有576%排出,DMA排出的量远远高于从食物中摄入的量,表明小鼠体内发生了砷的甲基化反应;在各脏器/组织中,仅在肠道中检出砷酸盐(AsⅤ),而蓄积含量(0.0054~0.0116μg·g^(-1))很低,在其余脏器/组织中的蓄积主要以砷胆碱(AsC)(0.0011~0.06124μg·g^(-1))、AsB(0.0031~0.0979μg·g^(-1))和DMA(0.0012~0.0070μg·g^(-1))为主,而这几种砷普遍被认为是低毒的。因此,正常膳食贝类引起的砷健康风险属于低关注级别。本研究有助于揭示人群日常摄食富砷贝类后砷在生物体内的累积分布及其代谢转化情况,同时也为人群正常摄食贝类的砷健康风险提供参考数据。The accumulation,metabolism,and transformation of arsenic in the organism were researched in a 14 d consecutive shellfish exposure experiment in mice.The results showed that after 14 d of dietary exposure,about 70%of the total arsenic ingested was finally excreted in feces.The accumulation of total arsenic in organs/tissues was as follows:stomach(0.26~0.54μg·g^(-1))>intestine(0.12~0.42μg·g^(-1))>spleen(0.031~0.162μg·g^(-1))>kidney(0.06~0.12μg·g^(-1))>liver(0.049~0.091μg·g^(-1)),with insignificant accumulation in the remaining organs/tissues(0.016~0.065μg·g^(-1)).About 46%of arsenobetaine(AsB)in food was excreted in feces,whereas about 576%of dimethylarsenic acid(DMA)was excreted.The content of DMA excreted was much higher than that ingested from the food,and thus arsenic methylation occurred in mice.Arsenate(AsⅤ)was only detected in the intestine,but the content(0.0054~0.0116μg·g^(-1))was very low.The accumulation of arsenic compounds in the remaining organs/tissues was mainly dominated by arseniccholine(AsC)(0.0011~0.06124μg·g^(-1)),AsB(0.0031~0.0979μg·g^(-1)),and DMA(0.0012~0.0070μg·g^(-1)),which were generally considered to be low-toxicity.Therefore,the health risk of arsenic caused through normal dietary shellfish is at a low level of concern.This study helps to reveal the cumulative distribution of arsenic and its metabolic transformation in organisms after ingestion of arsenic-rich shellfish,and also provides reference data on the health risk of arsenic in populations with normal consumption of shellfish.
