摘　　要：目的研究长链非编码核糖核酸(long non-coding RNA,lncRNA)HNF1A-AS1在胃癌患者组织内的表达及其与疾病预后的关联性。方法收集30例胃癌患者的癌组织及其癌旁组织样本,采用qRT-PCR技术,测定这些组织切片中lncRNA HNF1A-AS1的表达丰度。进一步剖析HNF1A-AS1表达水平与临床病理特征之间的内在联系,并采用Kaplan-Meier生存曲线来估算患者生存率,Cox比例风险回归模型对胃癌病患的预后情况进行分析。结果相较于癌旁组织,胃癌组织中HNF1A-AS1的表达量显升高(P<0.05)。在胃癌组织样本中,HNF1A-AS1的表达水平与肿瘤分期、分化程度、浸润深度以及淋巴结有无转移密切相关(P<0.05)。进一步分析表明,HNF1A-AS1的表达水平、肿瘤分期、分化程度以及淋巴结转移均为影响胃癌病患预后的独立因素(P<0.05)。结论胃癌组织内lncRNA HNF1A-AS1的表达水平与胃癌的临床病理特性及预后紧密相关,有望成为胃癌的潜在诊疗靶点。Objective To investigate the expression of long non-coding RNA(lncRNA)HNF1A-AS1 in gastric cancer tissues and its association with disease prognosis.Methods Samples of cancer tissues and adjacent normal tissues were collected from 30 gastric cancer patients.The expression abundance of lncRNA HNF1A-AS1 in these tissue sections was determined using quantitative reverse transcription polymerase chain reaction(qRT-PCR).Further was conclucted to explore the intrinsic relationship between HNF1A-AS1 expression level and clinicalpathological characteristics.Kaplan-Meier survival curves were used to estimate patients′survival rates,and Cox proportional hazards regression model was employed to analyze the prognosis of gastric cancer patients.Results HNF1A-AS1 expression in gastric cancer tissues was significantly increased compared to adjacent tissues(P<0.05).In gastric cancer tissue samples,the expression level of HNF1A-AS1 was closely related to tumor stage,differentiation degree,invasion depth and the presence or absence of lymph node metastasis(P<0.05).Further analysis showed that the expression level of HNF1A-AS1,tumor stage,differentiation degree and lymph node metastasis were independent factors affecting the prognosis of gastric cancer patients(P<0.05).Conclusion The expression level of lncRNA HNF1A-AS1 in the gastric cancer tissues is closely related to the clinical pathological characteristics and prognosis of gastric cancer,suggesting it may serve as a potential diagnosis and therapeutic target for gastric cancer.
