摘 要:本研究采用网络药理学与分子对接技术探究八角茴香活性成分缓解玉米赤霉烯酮致猪子宫内膜炎的作用机制。通过中药系统药理数据库与分析平台(TCMSP)数据库获取八角茴香主要有效活性成分及相关作用靶点,通过基因数据库(GeneCards)获取猪子宫内膜炎相关靶点;利用预测蛋白相互作用的数据库(String)建立蛋白互作(PPI)网络,结合生物网络绘图软件(Cytoscape)对靶点进一步筛选获取关键靶点,建立“有效成分-潜在作用靶点-通路”网络图在注释、可视化和综合发现数据库(DAVID)对可能作用靶点进行京都基因与基因组百科全书数据库(KEGG)信号通路和基因本体(GO)富集,选取排名靠前的有效活性成分与关键靶点经Autodock-Tools进行分子对接。结果表明:1)筛选出八角茴香的5个活性成分分别是白桦酸酯、木犀草素、山柰酚、儿茶素和槲皮素,缓解玉米赤霉烯酮子宫内膜炎的可能作用靶点25个。2)PPI网络分析结果发现,白细胞介素6(IL6)、白细胞介素1β(IL1β)、Toll样受体4(TLR4)和瘦素(LEP)是八角茴香缓解玉米赤霉烯酮致猪子宫内膜炎的关键靶点。3)KEGG信号通路富集发现,筛选的可能作用靶点主要参与炎症相关信号通路。4)分子对接预测结果表明,槲皮素和木犀草素与关键靶点均能稳结合。综上所述,八角茴香活性成分槲皮素和木犀草素可以通过炎症相关信号通路发挥抗炎和调节免疫作用,这为八角茴香缓解玉米赤霉烯酮致猪子宫内膜炎的应用提供了理论基础。The purpose of this study was to explore the mechanism of the active ingredients of star anise in alleviating zearalenone-induced porcine endometritis using network pharmacology and molecular docking technology.The active ingredients of star anise and related-targets,and the endometritis-related targets were obtained through the TCMSP database and GeneCards,respectively.The String database was used to establish a protein-protein interaction network(PPI).Subsequently,the key targets were further screened by Cytoscape software.The KEGG and GO enrichment of the possible targets were conducted in the DAVID database.The top effective active ingredients were selected,and the key targets underwent molecular docking through Autodock-Tools.Results showed as follows:1)five active ingredients including mairin,luteolin,kaempferolol,(+)-catechin and quercetin were screened,and 25 possible targets were selected to alleviate zearalenone-induced porcine endometritis.2)The PPI network analysis showed that interleukin 6(IL6),interleukin 1β(IL1β),Toll-like receptor 4(TLR4)and leptin(LEP)were the key targets of star anise in alleviating zearalenone-induced porcine endometritis.3)The enrichment of KEGG signaling pathway showed that the potential targets identified were primarily involved in inflammation-related signaling pathways.4)The molecular docking prediction results showed that both quercetin and luteolin could bind to key targets stably.In conclusion,the active ingredients of star anise,quercetin and luteolin,can exert anti-inflammatory and immunomodulating effects through inflammation-related signaling pathways.This provides a theoretical basis for the application of star anise in alleviating zearalenone-induced porcine endometritis.
