摘  要:目的:分析VI型胶原蛋白α1(collagen type VIalpha 1,Col6a1)在三阴性乳腺癌中的表达及其对三阴性乳腺癌细胞耐药性和干性的影响。方法:采用生物信息学方法分析Col6a1对三阴性乳腺癌患者预后的影响及在三阴性乳腺癌细胞中的表达以及与耐药相关蛋白ATP结合盒(ABC)超家族G成员2[TP-binding cassette(ABC)superfamily G member 2,ABCG2]的关系;采用免疫组化检测Col6a1在三阴性和非三阴性乳腺癌组织中的表达;采用MTT法检测Col6a1对三阴性乳腺癌细胞耐药性的影响;采用乳腺癌成球实验,检测Col6a1对三阴性乳腺癌细胞干性的影响。结果:Col6a1在三阴性乳腺癌患者中高表达,并与不良预后有关;Col6a1在三阴性乳腺癌组织及细胞系中过表达;Col6a1与ABCG2表达成正相关;Col6a1敲减后,三阴性乳腺癌细胞MDA-MB-231及Hs 578T对紫杉醇更敏感,并且会使这两种细胞的成球率降低。结论:Col6a1促进三阴性乳腺癌细胞耐药性及干性;Col6a1可作为三阴性乳腺癌治疗的一个潜在靶点。Objective:To analyze the expression of collagen type VI alpha 1(Col6a1)in triple-negative breast cancer and its effect on drug resistance and stemness of triple-negative breast cancer cells.Methods:Bioinformatics method analyzed the effect of Col6a1 on the prognosis in triple-negative breast cancer,the expression of Col6a1 in triple-negative breast cancer cells and the relationship with the drug resistance-related protein ATP-binding cassette(ABC)superfamily G member 2(ABCG2).Immunohistochemistry was used to detect the Col6a1 expression in triple-negative breast cancer tissues and non-triple-negative breast cancer tissues.The effect of Col6a1 on drug resistance in triple-negative breast cancer cells was examined by MTT assay.Mammosphere formation test was then used to evaluate the role of Col6a1 in triple-negative breast cell stemness.Results:Col6a1 was overexpressed in triple-negative breast cancer and was correlated with unfavorable prognosis.Col6a1 was overexpressed in triple-negative breast cancer tissues and cells.Col6a1 was positively correlated with ABCG2.Col6a1 knockdown enhanced the sensitivity of MDA-MB-231 cells and Hs 578T cells to paclitaxel(PTX)and impaired mammosphere formation capacity of these two cell lines.Conclusion:Col6a1 contributes to triple-negative breast cancer cells drug resistance and stemness,and can be a potential target for the treatment of triple-negative breast cancer.
