向日葵EPF基因家族鉴定及参与花发育的功能分析

摘  要:植物表皮模式因子EPF是一类广泛参与植物生长发育调节的分泌肽,其家族成员在向日葵(Helianthus annuus L.)花发育调控中的功能尚不清楚。基于基因组和转录组数据库,在向日葵中鉴定得到10个EPF家族基因,分属于4个亚家族。结果表明:其中,HaEPF1、HaEPF3、HaEPF4、HaEPF7、HaEPF9在向日葵花发育早期(St2、St3、St4)特异性高表达,而HaEPF5、HaEPF8、HaEPF10在花发育中后期(St5、St6、St7)特异性高表达。进一步分析发现,HaEPF1、HaEPF4、HaEPF7在野生型向日葵(WT)花分生组织转变和花器官发生(St2)过程中高表达,并随着花器官发育其表达水平逐渐降低。然而,10个EPF家族基因在向日葵花分生组织转变与花器官分化缺陷突变体Crazy Broccoli 1(cb1)中类花序结构发育过程中始终维持较为稳定的高表达水平,并与WT分生组织转变与花器官发生时期的表达水平相似。结果表明:HaEPF1、HaEPF4、HaEPF7可能在向日葵花分生组织转变和花器官发生过程中发挥作用;启动子顺式作用元件分析发现,HaEPF1、HaEPF4、HaEPF7在调控向日葵花分生组织转变和花器官发生过程中可能依赖于生长素、赤霉素、脱落酸信号。HaEPF1、HaEPF4、HaEPF7表达水平的精确调节对于向日葵花器官发生至关重要,持续异常地高表达,可能是导致cb1突变体出现花器官发育缺陷的重要原因之一。Epidermal patterning factor(EPF)is a type of secreted peptide involving in plant growth regulation,however,their functions in the progress of floral development in sunflower are still unclear.In this study,total 10 EPF genes were screened in sunflower based on the genome and transcriptome databases of sunflower,and they were categorized into four subfamilies.The results showed that among these genes,HaEPF1,HaEPF3,HaEPF4,HaEPF7,and HaEPF9 achieved their higher expression level at the early stages of floral development(St2,St3 and St4),while,HaEPF5,HaEPF8 and HaEPF10 expressed their optimal level at mid to late stages(St5,St6 and St7).Further analyses found that the expression of HaEPF1,HaEPF4 and HaEPF7 got to the peak at the stage of floral meristem transition and floral organogenesis(St2),and then gradually decreased with floral organs developing in normal sunflower,however,they kept on a stable higher expression during the whole progress of inflorescence development in Crazy Broccoli 1(cb1)sunflower with defects in floral meristem transition and floral organogenesis,with equal level to that of St2 in WT.It indicated that the HaEPF1,HaEPF4 and HaEPF7 genes could play an important role during floral meristem transition and floral organogenesis of sunflower.The results of cis acting elementanalysis showed that HaEPF1,HaEPF4 and HaEPF7 might regulate the progress of floral meristem transition and floral organogenesis depending on auxin,gibberellin,and abscisic acid signaling.The results suggested that in sunflower,the precise regulation of expression levels of HaEPF1,HaEPF4 and HaEPF7 was crucial for the floral organ development,and their sustained abnormal high-level expression might be one of the important reasons for the defects of floral organs of cb1 mutant.
