摘　　要：目的探讨Zeste增强子同源物2(enhancer of Zeste homolog 2,EZH2)在乳腺癌组织中的表达及对肿瘤进展和预后的影响。方法在癌症基因组图谱(the can-cer genome atlas,TCGA)数据库、基因表达综合数据库、欧洲基因组-表型档案数据库中获取乳腺癌组织和癌旁正常乳腺组织的转录组数据及患者的临床资料,利用TIMER 2.0平台分析EZH2的表达差异。在Kaplan-Meier Plotter数据库中获取乳腺癌患者的生存信息,比较EZH2低表达与高表达乳腺癌患者的总生存期、无复发生存期和无远处转移生存期。选取14只裸鼠,随机分为si-EZH2组和对照组,各7只。分别接种转染了EZH2敲降质粒和对照质粒的MCF7培养悬液。比较两组裸鼠接种MCF7细胞不同时间的体质量和肿瘤体积,第28天处死裸鼠剖取肿瘤,比较两组裸鼠的肿瘤质量。呈正态分布的计量资料以x¯±s表示,两组间比较采用两独立样本t检验;两组裸鼠不同时间体质量和肿瘤体积的比较采用重复测量的方差分析,两两比较采用Bonferroni检验检验。生存期的比较采用Log-rank检验。结果在TCGA数据库中共纳入乳腺癌组织1085份,癌旁正常乳腺组织291份,EZH2在乳腺癌组织中的表达高于癌旁正常乳腺组织(P<0.05)。在Kaplan-Meier Plotter数据库中,EZH2高表达组乳腺癌患者的总生存期、无复发生存期和无远处转移生存期均短于EZH2低表达组(P值分别为0.013、<0.001、<0.001)。接种MCF7培养悬液7 d后,两组裸鼠左侧背部均可见明显的皮下肿瘤。第1天,两组裸鼠体质量比较差异无统计学意义(P>0.05);第7、13、19、25、28天,两组裸鼠体质量和肿瘤体积均逐渐增加(体质量:F_(组内)=29.31、P<0.001,F_(组间)=234.32、P<0.001,F_(交互)=16.83、P<0.001;肿瘤体积:F_(组内)=34.00、P<0.001,F_(组间)=193.17、P<0.001,F_(交互)=35.61、P<0.001),且siEZH2组裸鼠体质量均高于对照组(均P<0.05),第19、25、28天,siEZH2组裸鼠肿瘤体积均小于对�ObjectiveTo investigate the expression characteristics of Zeste enhancer of Zeste homolog 2(EZH2)in breast cancer tissue and its influence on tumor progression and prognosis.MethodsTranscriptome data of breast cancer tissue and normal breast tissue adjacent to cancer as well as clinical data of patients were obtained from the cancer genome atlas(TCGA)database,gene expression comprehensive database and European genome phenotype archives database,and the difference of EZH2 expression was analyzed using TIMER 2.0 platform.The survival information of breast cancer patients was obtained from the Kaplan Meier Plotter database,and the overall survival time,relapse free survival time and distant metastasis free survival time of breast cancer patients with low EZH2 expression and high EZH2 expression were compared.Select 14 nude mice were selected and randomly divided into si-EZH2 group and control group,with 7 mice in each group.MCF7 culture suspensions transfected with EZH2 knockdown plasmid and control plasmid were inoculated for corresponding group.The body mass and tumor volume of two groups of nude mice inoculated with MCF7 cells were compared at different times.On the 28th day,the nude mice were euthanized and the tumors were dissected to compare the tumor mass of the two groups of nude mice.The normally distributed quantitative data was represented by x¯±s.Two independent sample t-tests were used for comparison between two groups,repeated measures ANOVA was used for comparison of body mass and tumor volume between two groups of nude mice at different times,and Bonferroni test was used for pairwise comparison.The comparison of survival rates was conducted using log rank test.ResultsA total of 1085 breast cancer tissues and 291 normal adjacent breast tissues were included in the TCGA database.EZH2 expression in breast cancer tissues was higher than that in normal adjacent breast tissues(P<0.05).In the Kaplan Meier Plotter database,the total survival time,relapse free survival time,and distant metastasis free survi
