威灵仙总多酚的提取及化学性质研究

摘  要:本研究以威灵仙为原料,采用溶剂提取法提取总多酚,并使用紫外分光光度计对威灵仙总多酚进行含量测定,通过单因素和正交实验优化提取工艺条件。对提取物质进行化学性质鉴别,明确提取物中含有多酚类物质,通过测定清除率检测其抗氧化活性。实验结果表明,最佳提取条件为浸提温度70℃、提取时间2.5h、料液比1∶20(g∶m L)和乙醇浓度70%,此时测得吸光度值为0.572,威灵仙总多酚的提取量为10.75mg·g^(-1),威灵仙中总多酚含量较高,通过颜色反应,初步鉴别提取物中含有酚羟基。This study used Climatidis radix as the raw material to extract total polyphenols using solvent extraction method,and determined the content of total polyphenols in Climatidis radix using UV spectrophotometer.The optimal extraction process conditions were optimized by single factor experiments and orthogonal experiments.The polyphenols were identified to exit in the extract.The experimental results showed that the extraction conditions of 70℃extraction temperature,2.5 hours extraction time,1∶20(g∶mL)solid-liquid ratio,and 70%ethanol concentration were the best.The absorbance value was 0.572 and an extraction amount was 10.75mg·g^(-1) under the condition.The total polyphenol content in Climatidis radix is relatively high.It was verified that there were phenolic hydroxyl groups in extract by color reaction tests.
