摘 要:采用串联质谱标签(TMT)标记的定量蛋白质组学技术,研究了-45℃超低温冻藏时间(0,3和6个月)对小黄鱼(Larimichthys polyactis)肌肉差异蛋白表达的影响。结果表明,与新鲜小黄鱼相比,在-45℃超低温冻藏3个月和6个月的小黄鱼肌肉中差异蛋白分别有71和104种,其中上调蛋白分别有13和40种,下调蛋白分别有58和64种。在冻藏6个月的小黄鱼肌肉中发生变化的蛋白质更多。此外,共筛选出15种差异蛋白,与小黄鱼的pH、持水力、挥发性盐基氮(TVBN)、硬度、弹性、羰基、巯基和Ca^(2+)-ATPase活性中的至少一个品质指标呈显著相关性,有望成为与冷冻小黄鱼品质相关的指示蛋白标志物。In order to investigate the effect of ultra-low temperature(-45℃)freezing time on the differential protein expression of small yellow croaker(Larimichthys polyactis)muscle,quantitative proteomics technology with TMT label was used.The results showed that compared with fresh yellow croaker,there were 71 and 104 different proteins in the muscle after 3 and 6 months of cryopreservation,including 13 and 40 up-regulated proteins and 58 and 64 downregulated proteins,respectively.More protein changes were found in the comparison between 0 and 6 months compared with the difference between 0 and 3 months.In addition,a total of 15 different proteins were selected,which were significantly correlated with at least one of the quality indexes of pH value,water-holding capacity,TVBN,hardness,springiness,as well as carbonyl content,total sulfhydryl and Ca~(2+)-ATPase activities of small yellow croaker(Larimichthys polyactis),and were expected to be indicator protein markers related to the quality of frozen small yellow croaker.
