摘 要:甲基化酶样16(Methyltransferase like 16,METTL16)作为N6-甲基腺苷(N6-methyladenosine,m6A)甲基转移酶家族重要成员,通过表观遗传修饰动态调控肿瘤发生发展。尽管METTL16在多种肿瘤中的重要性已得到证实,但目前对其在不同肿瘤类型中的分子机制异质性和功能多样性仍缺乏系统性的总结分析。本文基于最新研究进展,全面梳理了METTL16在肿瘤发生发展中的分子机制及其临床转化价值。研究表明,METTL16可通过m6A依赖性和非依赖性双重机制调控下游靶基因网络,进而影响肿瘤细胞周期进程、代谢重编程和铁死亡抵抗等关键生物学过程。研究显示,METTL16表达水平与一些肿瘤如胃癌、肝细胞癌、结直肠癌的预后明显相关。在治疗策略方面,靶向METTL16的干预措施已取得初步进展,包括通过对聚腺苷二磷酸核糖聚合酶抑制剂(Poly adp-ribose polymerase inhibitors,PARPi)增敏加强胰腺导管腺癌治疗效果,以及提高膀胱癌对顺铂化疗敏感性等。这些研究不仅揭示了METTL16作为肿瘤治疗关键靶点的重要价值,也为开发新型抗肿瘤策略提供了理论依据。As an important member of the N6-methyladenosine(m6A)methyltransferase family,methyltransferase like 16(METTL16)dynamically regulates tumorigenesis and development through epigenetic modification.Although the importance of METTL16 in various tumors has been confirmed,there is still a lack of systematic summary and analysis of its molecular mechanism heterogeneity and functional diversity in different tumor types.Based on the latest research progress,this article comprehensively reviews the molecular mechanism of METTL16 in tumorigenesis and development and its clinical transformation value.Studies have shown that METTL16 can requlate the downstream target gene network through m6A-dependent and m6A-independent mechanisms,thereby affecting key biological processes such as tumor cell cycle progression,metabolic reprogramming and ferroptosis resistance.Clinical transformation studies have shown that the expression level of METTL16 is significantly associated with the prognosis of some tumors such as gastric cancer,hepatocellular carcinoma,and colorectal cancer.In terms of treatment strategies,interventions targeting METTL16 have made initial progress,including enhancing the therapeutic effect of pancreatic ductal adenocarcinoma by sensitizing poly adp-ribose polymerase inhibitors(PARPi),and improving the sensitivity of bladder cancer to cisplatin chemotherapy.These results not only reveal the important value of METTL16 as a key target for tumor therapy,but also provide a theoretical basis for the development of new anti-tumor strategies.
