超高效液相色谱-高分辨质谱法同时测定刺梨原汁中2种三萜类化合物

摘  要:建立了刺梨原汁中2种三萜类化合物的超高效液相色谱-高分辨质谱检测方法。采用超声辅助溶剂提取刺梨原汁中三萜类化合物,以甲醇-0.1%甲酸水溶液作为流动相进行梯度洗脱,Hypersil GOLD色谱柱(150 mm×2.1 mm,1.9μm)分离,采用负离子扫描模式,全扫描/数据依赖性二级扫描(Full MS/ddMS^(2))模式采集数据,外标法定量。结果表明,2种三萜类化合物在各自线性范围内线性良好,相关系数(R^(2))大于0.995,加标回收率为89.5%~110.5%,相对标准偏差小于5.0%。运用该方法分析了不同品种、不同产区刺梨原汁中2种三萜类化合物的含量,结果表明不同品种刺梨原汁中刺梨苷含量具有显著差异,野鸦椿酸含量无显著差异;不同产区刺梨原汁中刺梨苷、野鸦椿酸含量均无显著差异。因此,建议若以刺梨苷作为功能成分开发产品,优先选择贵农5号品种刺梨。An ultra-high performance liquid chromatography-high resolution mass spectrometry method was established for the determination of two triterpenoids in Rosa roxburghii Tratt juice.The triterpenoids in Rosa roxburghii Tratt juice were extracted by ultrasonic-assisted solvent extraction,and separated by Hypersil GOLD column(150 mm×2.1 mm,1.9μm)with methanol-0.1%formic acid aqueous solution as the mobile phase under the gradient elution.The data were collected by Full MS/dd-MS^(2) in negative ion scanning mode,and quantitative analysis was realized by external standard method.The linearity of the two triterpenoids was good within the corresponding linear ranges,and the correlation coefficients(R^(2))were greater than 0.995.The recoveries of the spiked compounds were in the range of 89.5%-110.5%with the relative standard deviations less than 5.0%.The method was used to analyze the contents of two triterpenoids in Rosa roxburghii Tratt juice of different cultivars and different regions.The results showed that the contents of Kaji-ichigoside F1 in Rosa roxburghii Tratt juice of different cultivars had significant differences,while the contents of euscaphic acid had no significant differences.The contents of Kajiichigoside F1 and euscaphic acid had no significant differences in Rosa roxburghii Tratt juice from different regions.Therefore,it is suggested that if the product is developed with Kaji-ichigoside F1 as the functional component,Guinong 5 variety Rosa roxburghii Tratt is preferred.
