摘  要:目的探究松果菊苷(ECH)对慢性应激抑郁大鼠行为学及神经递质代谢的影响。方法通过不同类型的胁迫建立大鼠慢性不可预知温和应激抑郁模型(CUMS),通过腹腔注射松果菊苷对抑郁大鼠进行治疗。旷场实验(OFT)、强迫游泳测试(FST)、高架十字迷宫实验(EPM)被用来检测动物的行为学及抑郁状态。通过酶联免疫吸附(ELISA)试剂盒检测大鼠血清中5-羟色胺(5-HT)、5-羟吲哚乙酸(5-HIAA)、去甲肾上腺素(NE)、多巴胺(DA)水平,采取蛋白免疫印迹法(Western blot)对大鼠海马组织BDNF蛋白水平进行检测。结果ECH降低CUMS引起的大鼠体质量的降低,减少CUMS诱导的大鼠抑郁行为,升高单胺能神经递质水平并且部分恢复大鼠海马组织BDNF蛋白水平。结论ECH能够促进神经递质代谢并抑制大鼠抑郁行为。Objective To explore the effect of echinacoside(ECH)on behavior and neurotransmitters metabolism in chronic stress-induced depression rats.Methods A chronic unpredictable mild stress(CUMS)-induced depression rat model was established by different stress,and the depression rats received intraperitoneal injection of ECH.The open field test(OFT),forced swim test(FST)and elevated plus maze(EPM)test were performed to assess the behavior and depression in the rats.Enzyme-linked immunosorbent assay(ELISA)was employed for the detection of the serum levels of 5-hydroxytryptamine(5-HT),5-hydroxyindoleacetic acid(5-HIAA),norepinephrine(NE),dopamine(DA),Western blot(WB)were conducted to evaluated brain-derived neurotrophic factor(BDNF)protein levels in rat hippocampus.Results ECH downregulated CUMS-induced weight less and depressive-like behaviors,upregulated monoamine neurotransmitter levels,and partially restored BDNF protein levels in rat hippocampus.Conclusion ECH can promote neurotransmitters metabolism and inhibite depressive-like behaviors in rats.
