摘  要:目的探究瞬时受体电位melastatin2蛋白(TRPM2)在前列腺癌中的表达及其与患者预后和免疫浸润的相关性。方法使用TIMER数据库研究前列腺癌肿瘤与癌旁组织之间的TRPM2表达水平。评估TRPM2表达水平的预后价值;评估前列腺癌易感基因在不同突变状态下的TRPM2基因的表达水平;分析前列腺癌中TRPM2表达与肿瘤微环境的相关性。通过荧光定量聚合酶链反应验证前列腺癌组织中TRPM2 m RNA表达水平,通过Elisa验证前列腺癌血浆中前列腺特异性抗原(PSA)和TRPM2蛋白表达水平及两者的相关性。通过Elisa验证前列腺癌患者手术前后血浆中TRPM2蛋白表达水平。结果前列腺癌组织中TRPM2表达水平显著高于对应的癌旁组织;TRPM2表达水平与前列腺癌患者的不良预后无显著相关性,与前列腺癌易感基因具有一定相关性。TRPM2表达与调节性T细胞、非调节性CD4+T细胞、CD8+T细胞、B细胞、NK细胞、中性粒细胞、单核细胞、癌症相关成纤维细胞、巨噬细胞、内皮细胞、树突状细胞浸润呈正相关,与肥大细胞、CD4+T细胞浸润呈负相关。前列腺癌中TRPM2 m RNA表达水平显著高于对应的癌旁组织(P<0.001),前列腺癌血浆中PSA与TRPM2表达水平与肿瘤临床分期呈正相关(P<0.001),且二者具有一定相关性。前列腺癌患者术后血浆TRPM2表达水平显著低于术前(P<0.001)。结论TRPM2在前列腺癌组织中高表达,与患者不良预后无显著相关性,但与前列腺癌易感基因相关,与前列腺癌免疫浸润相关,是潜在的预后生物标志物和免疫靶点。Objective To explore the expression of transient receptor potential melastatin 2(TRPM2)in prostate cancer and its correlation with the prognosis and immune infiltration.Methods The TIMER database was used to investigate the expression levels of TRPM2 between prostate cancer tumors and adjacent normal tissues.The prognostic value of TRPM2 expression levels was evaluated;the expression levels of the TRPM2 gene under different mutation states of prostate cancer susceptibility genes was assessed;the correlation between TRPM2 expression and the tumor microenvironment in prostate cancer was analyzed.The expression level of TRPM2 mRNA in prostate cancer tissues was verified by RT-qPCR.The expression levels of prostate-specific antigen(PSA)and TRPM2 protein in prostate cancer plasma,as well as the correlation between them,were verified by ELISA.The expression level of TRPM2 protein in the plasma of patients with prostate cancer before and after surgery was verified by ELISA.Results The expression level of TRPM2 in prostate cancer tissues was significantly higher than that in the corresponding adjacent tissues;there was no significant correlation between TRPM2 expression level and the poor prognosis of prostate cancer patients;the expression level of TRPM2 had a certain correlation with prostate cancer susceptibility genes.TRPM2 expression was positively correlated with the infiltration of regulatory T cells,non-regulatory CD4+T cells,CD8+T cells,B cells,NK cells,neutrophils,monocytes,cancer-associated fibroblasts,macrophages,endothelial cells,and dendritic cells,and negatively correlated with the infiltration of mast cells and CD4+T cells.The mRNA expression of TRPM2 in prostate cancer was significantly higher than that in the corresponding adjacent tissues(P<0.001);the expression levels of PSA and TRPM2 in prostate cancer plasma were positively correlated with the tumor clinical stage(P<0.001),and there was a certain correlation between PSA and TRPM2.The expression level of TRPM2 in the plasma of prostate cancer patient
