摘  要:目的探究甲状腺乳头状癌(PTC)组织中长链非编码RNA(LncRNA)核富含丰富的转录本1(NEAT1)及微小RNA(miR)-34a的表达量及临床意义。方法选取2016年11月至2018年6月商洛市中心医院收治的89例PTC患者,术中收集PTC组织及癌旁组织(PT)。实时荧光定量逆转录聚合酶链反应检测LncRNA NEAT1、miR-34a表达量。Pearson法分析PTC组织中LncRNA NEAT1和miR-34a表达量的相关性。以Kaplan-Meier生存曲线分析LncRNA NEAT1和miR-34a相对表达量与PTC预后的关系,并以Cox回归模型分析影响PTC患者预后的因素。结果PTC组织中LncRNA NEAT1相对表达量(4.19±0.90)较PT(1.36±0.34)高,miR-34a相对表达量(0.43±0.12)较PT(1.33±0.36)低(P<0.05)。LncRNA NEAT1和miR-34a表达量在癌组织分化程度方面存在统计学差异(P<0.05),且miR-34a表达量在淋巴结转移方面亦存在统计学差异(P<0.05)。PTC组织中LncRNA NEAT1和miR-34a表达量呈负相关(r=-0.516,P<0.05)。高表达LncRNA NEAT1患者的3年生存率低于低表达患者(P<0.05),高表达miR-34a患者的3年生存率高于低表达患者(P<0.05)。低表达miR-34a、高表达LncRNA NEAT1及低分化是影响PTC患者预后的独立危险因素(P<0.05)。结论PTC组织中LncRNA NEAT1高表达,miR-34a低表达,且二者呈现负相关,均有望成为新的PTC预后评估的肿瘤标志物。Objective To investigate the expression levels and clinical significance of long non-coding RNA(LncRNA)nuclear-enriched abundant transcript 1(NEAT1)and microRNA(miR)-34a in papillary thyroid cancer(PTC)tissues.Methods A total of 89 patients with PTC who were admitted to our hospital from November 2016 to June 2018 were selected.Both PTC tissues and paracancerous tissues(PT)were collected during surgery.Real-time fluorescence quantitative RTPCR was used to detect the expression levels of LncRNA NEAT1 and miR-34a in PTC tissues.Pearson's correlation analysis was performed to analyze the correlation between LncRNA NEAT1 and miR-34a expression levels.The Kaplan-Meier survival curve was used to analyze the relationship between the relative expression of LncRNA NEAT1,miR-34a and PTC prognosis.Cox regression models were used to analyze the factors affecting the prognosis of PTC patients.Results The relative expression level of LncRNA NEAT1 in PTC tissues(4.19±0.90)was significantly higher than that in PT(1.36±0.34),while the miR-34a expression(0.43±0.12)was lower than that in PT(1.33±0.36)(P<0.05).There were significant differences in the expression quantity of LncRNA NEAT1 and miR-34a in the differentiation degree of cancer tissues,and there were also significant differences in the expression quantity of miR-34a in lymph node metastasis(P<0.05).The expression levels of LncRNA NEAT1 and miR-34a in PTC tissues are negatively correlated(r=-0.516,P<0.05).The high expression of LncRNA NEAT1 of patients 3-year survival rate was lower than that of patients with low expression(P<0.05),and the high expression of miR-34a of patients 3-year survival rate was lower than that of patients with low expression(P<0.05).Low expression of miR-34a,high expression of LncRNA NEAT1,and low differentiation are independent risk factors affecting the prognosis of PTC patients(P<0.05).Conclusion LncRNA NEAT1 is highly expressed in PTC tissues,while miR-34a is low expressed,showing a negative correlation.Both molecules may serve as novel biom
