摘  要:目的探讨丹皮酚治疗宫颈癌的作用机制。方法通过TCMSP、PubChem、STITCH和BATMAN-TCM数据库筛选丹皮酚相关靶点,结合Gene Cards和OMIM数据库筛选宫颈癌相关靶点,筛选出交集靶点;经STRING构建蛋白质—蛋白质相互作用(PPI)网络及Cytoscape拓扑分析筛选核心靶点;利用仙桃学术平台进行基因本体(GO)功能富集分析与京都基因和基因组百科全书(KEGG)通路富集分析。结果筛选得到丹皮酚作用靶点109个,宫颈癌作用靶点1372个,丹皮酚治疗宫颈癌潜在靶点28个。PPI筛选出苏氨酸激酶1(AKT1)、肿瘤坏死因子(TNF)、半胱天冬蛋白酶-3(CASP3)和B淋巴细胞瘤-2基因(BCL2)为丹皮酚治疗宫颈癌关键靶点。GO功能富集分析显示,丹皮酚主要靠转录共激活因子结合、外膜、外源性凋亡信号通路等过程发挥作用;KEGG通路富集分析涉及PI3K-AKT及NF-κB信号通路。结论丹皮酚可能通过靶向调控AKT1、TNF、CASP3和BCL2介导的PI3K-AKT及NF-κB信号通路治疗宫颈癌。Objective To analyze the mechanism of action of paeonol in cervical cancer treatment.Methods Pae-onol-related targets were screened through databases including TCMSP,PubChem,STITCH and BATMAN-TCM.Cervical cancer-related targets were identified using Gene Cards and OMIM databases,followed by screening for intersection tar-gets.A protein-protein interaction(PPI)network was constructed via STRING,and core targets were selected through to-pological analysis using Cytoscape.Gene Ontology(GO)functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis were performed using Xiantao Academic.Results A total of 109 paeonol-related targets and 1372 cervical cancer-related targets were screened,with 28 potential anti-cervical cancer tar-gets obtained.PPI analysis identified core targets including AKT1,TNF,CASP3,and BCL2.GO functional enrichment analysis showed that paeonol primarily acts through processes such as transcription coactivator binding,outer membrane,and extrinsic apoptotic signaling pathway.KEGG pathway enrichment analysis involved the PI3K-AKT and NF-κB signal-ing pathways.Conclusion Paeonol may treat cervical cancercervical cancer by targeting and regulating AKT1,TNF,CASP3 and BCL2 to mediate the PI3K-AKT and NF-κB signaling pathways.
