胃癌患者血清miRNA-132和miRNA-345表达与肿瘤标志物及预后关系研究

摘　　要：目的探讨胃癌患者血清微小RNA-132(miRNA-132)和微小RNA-345(miRNA-345)表达与肿瘤标志物及预后的关系。方法选择2020年1月至2022年1月于该院治疗的103例胃癌患者作为研究组;选择同期于该院治疗的82例胃部良性疾病患者作为良性组;另选择同期于该院体检的61例健康体检者作为对照组。采用实时荧光定量聚合酶链反应(qPCR)法测定miRNA-132和miRNA-345表达;采用电化学发光法测定癌胚抗原(CEA)、糖类蛋白抗原199(CA199)和糖蛋白抗原724(CA724)水平。所有患者随访至2024年6月,统计胃癌患者总生存期(OS),通过门诊或电话随访。结果研究组血清miRNA-132相对表达量高于良性组和对照组(P<0.05),而miRNA-345相对表达量低于良性组和对照组(P<0.05)。研究组血清CEA、CA199和CA724水平高于良性组和对照组(P<0.05)。经Pearson分析显示,miRNA-132与CEA、CA199和CA724呈正相关(P<0.05),而miRNA-345与CEA、CA199和CA724呈负相关(P<0.05)。miRNA-132相对表达量≤2.76患者OS优于miRNA-132相对表达量>2.76患者(P<0.05)。miRNA-132相对表达量>0.37患者OS优于miRNA-132相对表达量≤0.37患者(P<0.05)。miRNA-132高表达和miRNA-345低表达为影响胃癌患者预后危险因素(P<0.05)。结论胃癌患者miRNA-132呈高表达而miRNA-345呈低表达,且miRNA-132和miRNA-345表达与肿瘤标志物和预后密切相关,值得临床借鉴。Objective To investigate the relationship between serum microRNA-132(miRNA-132)and microRNA-345(miRNA-345)expression,tumor markers and prognosis in gastric cance r patients.Methods A total of 103 gastric cancer patients from the hospital from January 2020 to January 2022 were selected as study group.82 patients with benign gastric diseases in the hospital during the same period were selected as the benign group.Another 61 healthy individuals who underwent physical examinations in the hospital during the same period were selected as control group.Real-time polymerase chain reaction(qPCR)was used to measure the expression of miRNA-132 and miRNA-345.The levels of carcinoembryonic antigen(CEA),carbohydrate antigen 199(CA199)and glycoprotein antigen 724(CA724)were measured using electrochemiluminescence.All patients were followed up until June 2024,and the overall survival(OS)of gastric cancer patients was calculated through outpatient or telephone follow-up.Results The relative expression level of miRNA-132 in the serum of the study group was higher than that of the benign group and the control group(P<0.05),while the relative expression level of miRNA-345 was lower than that of the benign group and the control group(P<0.05).The serum levels of CEA,CA199,and CA724 in the study group were higher than those in the benign group and the control group(P<0.05).Pearson analysis showed that miRNA-132 was positively correlated with CEA,CA199,and CA724(P<0.05),while miRNA-345 was negatively correlated with CEA,CA199,and CA724(P<0.05).Patients with a relative expression level of miRNA-132≤2.76 had better OS than those with a relative expression level of miRNA-132>2.76(P<0.05).Patients with a relative expression level of miRNA-132 greater than 0.37 have better OS than those with a relative expression level of miRNA-132≤0.37(P<0.05).High expression of miRNA-132 and low expression of miRNA-345 were risk factors affecting the prognosis of gastric cancer patients(P<0.05).Conclusion Patients with gastric cancer have high expressi
