摘  要:制备一种植物乳杆菌(Lactobacillus plantarum)能力验证样品,并进行均匀性和稳定性的研究。首先制备植物乳杆菌的能力验证样品,经过冻干保护剂的单因素实验以及正交实验,得出冻干保护剂最佳配方,然后制备的能力验证样品按照CNAS-GL003:2018《能力验证样品均匀性和稳定性评价指南》的要求进行均匀性和稳定性检验。得出植物乳杆菌最佳冻干保护剂配方为15%脱脂乳粉、15%海藻糖、3%谷氨酸钠、2%抗坏血酸,能力验证样品的均匀性和稳定性能够满足能力验证样品的要求。能力验证样品均匀性好、稳定性佳,能够准确地考查乳酸菌微生物检验人员的检测水平,为国内微生物能力验证水平的提高奠定了基础。To prepare Lactobacillus plantarum proficiency testing samples with sufficient homogeneity and good stability.Firstly,the proficiency testing samples of Lactobacillus plantarum was prepared,and the optimal formula of the lyophilized protective agent was obtained through the single factor experiment and orthogonal experiment of the lyophilized protective agent.Then,the prepared proficiency testing samples was tested for its homogeneity and stability according to the requirements of CNAS-GL003:2018“Guidance on Evaluating the Homogeneity and Stability of Samples Used for Proficiency Testing”.It was concluded that the optimal formula of lyophilized protective agent for Lactobacillus plantarum was 15%skim milk powder,15%trehalose,3%sodium glutamate and 2%ascorbic acid.The homogeneity and stability of the sample could meet the requirements of the sample for proficiency testing.proficiency testing samples has good homogeneity and stability,and can accurately examine the detection level of lactobacillus microbial inspectors,which lays a foundation for the improvement of domestic microbial ability verification level.
