摘 要:【目的】研究贝莱斯芽孢杆菌(Bacillus velezensis)G18对枸杞根腐病的抗病性及其对枸杞的促生作用,从活性氧(reactive oxygen species,ROS)积累和相关防御酶活性变化的角度,揭示贝莱斯芽孢杆菌(B.velezensis)G18对枸杞根腐病病原菌腐皮镰刀菌(Fusarium solani)的抑菌机理,为枸杞根腐病的生物防治提供理论依据。【方法】以腐皮镰刀菌和贝莱斯芽孢杆菌为供试菌株,以1年生‘宁杞7号’健康植株为材料,采用盆栽试验设置单接种贝莱斯芽孢杆菌G18、单接种腐皮镰刀菌、接种贝莱斯芽孢杆菌G187 d后再接种腐皮镰刀菌、接种腐皮镰刀菌发病后再接种贝莱斯芽孢杆菌G18、接种腐皮镰刀菌发病后灌根2.5%咪鲜胺以及无菌水对照6个处理,测定不同处理枸杞的形态指标、生理指标,并计算发病率、病情指数和相对防效;采用试剂盒测定贝莱斯芽孢杆菌G18发酵滤液对腐皮镰刀菌活性氧含量和相关防御酶活性的影响。【结果】与单接种腐皮镰刀菌相比,接种贝莱斯芽孢杆菌G18可显著降低枸杞根腐病的发病率和病情指数,贝莱斯芽孢杆菌G18对腐皮镰刀菌的保护防效和治疗防效分别为62.3%和54.4%。与CK相比,贝莱斯芽孢杆菌G18显著促进了枸杞的生长(包括株高、茎粗、叶片干鲜质量、叶长、叶宽、叶面积和叶形指数),叶片中叶绿素、可溶性蛋白、可溶性糖、硝态氮、无机磷含量均显著增加;贝莱斯芽孢杆菌G18发酵滤液显著提高了腐皮镰刀菌的过氧化氢(H_(2)O_(2))和超氧阴离子(O_(2)^(-·))含量,羟自由基(·OH)清除率,以及谷胱甘肽过氧化物酶(GPX)和抗坏血酸过氧化物酶(APX)活性;显著降低了腐皮镰刀菌的超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)的活性。【结论】贝莱斯芽孢杆菌G18是具有较高开发价值的生防细菌,可显著促进枸杞生长,并大幅提高其抗根腐病的能力。【Objective】The resistance of Bacillus velezensis to root rot in Lycium barbarum and its growth-promoting effects on L.barbarum were studied.The antifungal mechanism of B.velezensis on Fusarium solani,the pathogen of root rot in L.barbarum,was revealed from the perspective of reactive oxygen species(ROS)accumulation and related defense enzyme activities,which provided a theoretical basis for the biological control of root rot in L.barbarum.【Method】One-year-old healthy“Ningqi 7”plants were used as experimental materials,and F.solani and B.velezensis were selected as the test strains.A pot experiment was conducted with six treatment groups as follows:single inoculation with B.velezensis G18,single inoculation with F.solani,inoculation with B.velezensis G18,followed by inoculation with F.solani 7 days later,sterile water control group(no inoculation with either strain),inoculation with Fusarium solani followed by inoculation with Bacillus velezensis G18.The morphological and physiological indexes of L.barbarum treated with different treatments were measured,and subsequently,the incidence rate,disease index,and relative control effect were calculated.The kit was used to determine the effects of B.velezensis G18 fermentation filtrate treatment on the active oxygen content and related defense enzyme activity in F.solani.【Result】Compared with single inoculation of F.solani,inoculation of B.velezensis G18 could significantly reduce the incidence and disease index of L.barbarum root rot.The protective and therapeutic efficacy of Bacillus subtilis G18 against Fusarium solani were 62.3%and 54.4%,respectively.Compared with the control(CK),B.velezensis G18 significantly promoted the growth of L.barbarum(including plant height,stem diameter,leaf area,dry and fresh weight,length,width,area and shape index),and the contents of chlorophyll,soluble protein,soluble sugar,nitrate nitrogen and inorganic phosphorus in leaves were also significantly increased.The fermentation filtrate of B.velezensis G18 significantly in
