摘 要:目的研究反式没药甾酮(Z-GS)在体内外对胃癌细胞BGC823增殖、凋亡的影响,及其对环氧合酶-2(COX-2)蛋白表达的影响。方法选取COX-2高表达的人胃腺癌细胞株BGC823,用不同浓度的Z-GS(0、4、8、12、16、20、24μg/ml)分别作用于胃癌细胞12、24、48 h,MTT法观察Z-GS对胃癌细胞增殖的影响。细胞分为对照组(0μg/ml Z-GS),Z-GS低、中、高浓度组(8、12、16μg/ml Z-GS),Z-GS干预胃癌细胞24 h。通过Hoechst 33342染色法观察Z-GS作用胃癌后细胞凋亡的形态变化,流式细胞术检测细胞的凋亡率。Western blot法检测COX-2、Bcl-2、Bax蛋白表达水平。选取12只无胸腺裸鼠构建裸鼠胃肿瘤模型,按照随机数字表法将其分为动物实验组、动物对照组,各6只。动物实验组以Z-GS灌胃处理,动物对照组以生理盐水灌胃处理的裸鼠。观察裸鼠的肿瘤生长和体重变化情况,检测裸鼠胃肿瘤组织中COX-2蛋白表达水平。结果各浓度Z-GS(4、8、12、16、20、24μg/ml)处理后的细胞增殖率与0μg/ml Z-GS比较,差异有统计学意义(P<0.05)。Z-GS作用后,与对照组比较,Z-GS低、中、高浓度组凋亡细胞呈现出典型的凋亡形态学改变。与对照组比较,Z-GS低、中、高浓度组细胞凋亡率升高(P<0.05);Z-GS中、高浓度组细胞凋亡率高于Z-GS低浓度组,Z-GS中浓度组细胞凋亡率高于Z-GS低浓度组(P<0.05)。与对照组比较,Z-GS低、中、高浓度组胃癌细胞中,Bcl-2、COX-2表达水平降低,Bax表达水平升高(P<0.05);与Z-GS低浓度组比较,Z-GS中、高浓度组胃癌细胞中,Bcl-2、COX-2表达水平降低,Bax表达水平升高(P<0.05);与Z-GS中浓度组比较,Z-GS高浓度组胃癌细胞中,Bcl-2、COX-2表达水平降低,Bax表达水平升高(P<0.05)。动物实验组皮下移植瘤体积小于动物对照组(P<0.05)。动物实验组和动物对照组体重比较,差异无统计学意义(P>0.05)。与动物对照组比较,动物实验组肿瘤组织的COX-2蛋白表达水平降低(P<0.Objective To investigate the effects of Z-Guggulsterone(Z-GS)on the proliferation and apoptosis of gastric cancer cells BGC823 in vitro and in vivo,and its effect on the expression of cyclooxygenase-2(COX-2)protein.Methods Human gastric cancer cell line BGC823 with high expression of COX-2 was selected,and gastric cancer cells were treated with different concentrations of Z-GS(0,4,8,12,16,20μg/ml and 24μg/ml)for 12,24,48 h,respectively.MTT assay was used to observe the effect of Z-GS on the proliferation of gastric cancer cells.The cells were divided into con-trol group(0μg/ml Z-GS),Z-GS low,medium,and high concentration groups(8,12,16μg/ml Z-GS),and Z-GS intervened gastric cancer cells for 24 h.Hoechst 33342 staining was used to observe the morphological changes of apoptosis after Z-GS treatment of gastric cancer,and flow cytometry was used to detect the apoptosis rate of cells.The protein expression levels of COX-2,Bcl-2,and Bax were detected by Western blot.Twelve athymic nude mice were selected to construct gastric tumor model in nude mice.According to the random number table method,they were divided into animal experimental group and animal control group,with six mice in each group.The animal experimental group was treated with Z-GS by gavage,and the animal control group was treated with normal saline by gavage.The tumor growth and body weight of nude mice were observed,and the protein level of COX-2 in gastric tumor tissues of nude mice was detected.Results The cell proliferation rate of each concentration of Z-GS(4,8,12,16,20,24μg/ml)were statistically significant compared with 0μg/ml of Z-GS(P<0.05).After Z-GS treatment,compared with the control group,the apoptotic cells in the Z-GS low,medium,and high concentration groups showed typical apoptotic morphological changes.Compared with the control group,the apoptosis rate of Z-GS low,medium,and high concentration groups increased(P<0.05).The apoptosis rate of Z-GS medium and high concentration groups was higher than that of Z-GS low concentration group
