摘 要:目的:利用生物信息学研究氨酰-tRNA合成酶1(AARS1)在结直肠癌(CRC)中的表达和预后及其与免疫浸润的关系,探索相关分子机制。方法:通过TCGA数据集和HPA数据库验证AARS1在CRC中的表达。利用GEPIA2数据库分析AARS1转录本表达。借助Kaplan-Meier Plotter网站分析AARS1表达与预后的关系。使用MethSurv数据库分析AARS1的CpG岛甲基化。利用GSCA和GEPIA2数据库分析AARS1表达与免疫浸润和免疫检查点的关系。依托TCGA数据集筛选AARS1相关蛋白并进行GO、KEGG和GSEA富集分析。利用STRING和GeneMANIA数据库构建AARS1蛋白互作网络。结果:AARS1在CRC中高表达并与患者预后呈负相关,此外其甲基化水平降低。AARS1可通过调控免疫分子和PD-1等免疫检查点影响CRC预后,此外,AARS1相关蛋白参与调控rRNA表达等生物活动,从而影响结直肠癌的发生与发展。结论:AARS1作为预后生物标志物影响CRC的免疫浸润,可作为其诊断和治疗的有效靶点。Objective:To investigate the expression and prognosis of aminoacyl-tRNA synthetase 1(AARS1)in colorectal cancer and its relationship with immune infiltration using bioinformatics,and explore the relevant molecular mechanisms.Methods:The expression of AARS1 in CRC was investigated using the TCGA dataset and HPA database.The expression of the AARS1 transcript was analyzed using the GEPIA2 database.The relationship between AARS1 expression and prognosis was analyzed using the Kaplan-Meier Plotter website.The CpG island methylation of AARS1 was analyzed using the MethSurv database.The relationship between AARS1 expression,immune infiltration,and immune checkpoint was analyzed using the GSCA and GEPIA2 databases.AARS1-related proteins were screened based on the TCGA dataset,and GO,KEGG,and GSEA enrichment analyses were performed.An AARS1 protein interaction network was constructed using the STRING and GeneMANIA databases.Results:AARS1 was found to be highly expressed in CRC and was negatively correlated with patient prognosis,with decreased methylation levels observed.AARS1 influenced the prognosis of CRC by regulating immune molecules and immune checkpoints such as PD-1.Additionally,the proteins related to AARS1 was involved in biological activities,including the regulation of rRNA expression,thereby affecting the occurrence and progression of CRC.Conclusion:AARS1,as a prognostic biomarker affecting immune infiltration in CRC,can serve as an effective target for its diagnosis and treatment.
