摘  要:抑郁症是一种异质性精神疾病,遗传易感性和环境因素之间的相互作用在其发病过程中扮演着关键角色。该病的具体机制仍需深入研究与阐释,但目前已有广泛共识认为,表观遗传标记对其作用机制具有重要影响。脑源性神经营养因子(brain-derived neurotrophic factor,BDNF)的DNA甲基化,不仅被视为一种有前景的病理学表观遗传生物标志物,还可能有助于预测抗抑郁药的疗效。该文综述了BDNF的基因结构及其DNA甲基化调控机制,同时分析了抑郁症患者及动物模型中该因子DNA甲基化的变化情况,旨在为临床研究提供新的思路和理论支撑。Depression is a heterogeneous mental disorder in which the interaction between genetic susceptibility and environmental factors plays a key role in its pathogenesis.Although the specific mechanisms of the disease still need to be thoroughly studied and elucidated,there is now a broad consensus that epigenetic markers have a central influence on its mechanism of action.DNA methylation of brain-derived neurotrophic factor(BDNF)is not only regarded as a promising epigenetic biomarker of pathology,but may also help predict the efficacy of antidepressants.In this paper we reviewed the gene structure of BDNF and its DNA methylation regulation mechanism,and also analyzed the changes of DNA methylation of this factor in depression patients and animal models,aiming to provide new ideas and theoretical support for clinical research.
