摘 要:目的探讨清解化攻方调控PI3K/AKT/NF-κB信号通路对急性胰腺炎(acute pancreatitis,AP)小鼠炎症反应的治疗作用及机制。方法24只小鼠随机分为空白组、模型组、乌司他丁组、清解化攻方组,每组6只。采用雨蛙素制备AP模型。ELISA法检测血清α-AMS、PNLP、IL-1β、IL-6、IL-8、IL-18、TNF-α水平;采用HE染色检测胰腺病理;免疫组化、Western blot和RT-qPCR检测PI3K、AKT、NF-κB相关蛋白和mRNA表达。结果与空白组比较,模型组胰腺明显病理损伤,血清α-AMS、PNLP、IL-1β、IL-6、IL-8、IL-18、TNF-α含量明显升高(P<0.01),PI3K、AKT、NF-κB相关蛋白及mRNA表达水平明显升高(P<0.01);与模型组对比,清解化攻方组和乌司他丁组均可改善AP小鼠胰腺组织病理变化,降低血清α-AMS、PNLP、IL-1β、IL-6、IL-8、IL-18、TNF-α含量,下调胰腺PI3K、AKT、NF-κB相关蛋白及mRNA表达水平(P<0.05或P<0.01)。结论清解化攻方可能通过调控PI3K/AKT/NF-κB信号通路的表达,抑制炎症反应,保护胰腺组织。Aim To explore the therapeutic effect and mechanism of Qingjie Huagong decoction in modulating PI3K/AKT/NF-κB signaling pathway in inflammatory response of acute pancreatitis(AP)mice.Methods Twenty-four mice were randomly divided into Blank group,Model group,Ustekin group,and Qingjie Huagong decoction group,with six mice in each group.The AP model was prepared by using rain frogin.Serumα-AMS,PNLP,IL-1β,IL-6,IL-8,IL-18,and TNF-αlevels were detected by ELISA;the pancreatic pathology was detected by HE staining;the expressions of PI3K,AKT,and NF-κB-related proteins and mRNAs were detected by immunohistochemistry,Western blot,and RT-qPCR.Results Compared with the blank group,the model group showed obvious pathological damage to the pancreas,with significantly higher serumα-AMS,PNLP,IL-1β,IL-6,IL-8,IL-18,and TNF-αlevels(P<0.01),and significantly higher levels of PI3K,AKT,and NF-κB-related proteins and mRNA expression(P<0.01).Compared with the model group,both the Qingjie Huagong decoction group and the ustekin group improved the histopathological changes in the pancreas of AP mice,decreased the serumα-AMS,PNLP,IL-1β,IL-6,IL-8,IL-18,and TNF-αlevels,and down-regulated the expression levels of pancreatic PI3K,AKT,NF-κB-related proteins and mRNA(P<0.05 or P<0.01).Conclusion Qingjie Huagong decoction may inhibit the inflammatory response and protect pancreatic tissues by regulating the expression of PI3K/AKT/NF-κB signaling pathway.
