摘 要:目的基于Nrf2/HO-1信号通路探讨真武汤对脂多糖(lipopolysaccharides,LPS)诱导的人肾小球上皮细胞(human glomerular epithelial cells,HGEC)炎症、氧化应激和凋亡的影响及作用机制。方法用LPS(1.0 mg·L-1)处理HGEC 24 h构建氧化损伤模型,在此基础上加入2.5%、5%、10%真武汤含药血清联合处理为真武汤低、中、高剂量组,并设正常组;采用MTT法和LDH法检测细胞活性的变化;采用ELISA法检测各组细胞中TNF-α、IL-6、IL-10、SOD、CAT、GSH-Px、ROS和MDA的含量;采用流式细胞术检测各组细胞凋亡情况;分别采用RT-qPCR和Western blot检测Bax、Bcl-2、caspase-3、caspase-9及Nrf2/HO-1通路的mRNA和蛋白表达情况。结果与正常组比较,模型组HGEC炎性因子含量升高,氧化应激反应和细胞凋亡加重;经真武汤各剂量组干预后,HGEC炎性水平降低,氧化应激和细胞凋亡得到有效改善,同时上调了Nrf2/HO-1信号通路的mRNA和蛋白表达水平。结论真武汤能够保护HGEC减缓LPS诱导的炎症和氧化损伤,改善细胞凋亡,其作用机制可能与其激活Nrf2/HO-1信号通路有关。Aim To investigate the effect of Zhenwu decoction on inflammation,oxidative stress and apoptosis of human glomerular epithelial cells(HGEC)induced by lipopolysaccharide(LPS)based on Nrf2/HO-1 signaling pathway,and to explore the underlying mechanism.Methods HGEC were treated with LPS(1.0 mg·L-1)for 24 h to construct an oxidative damage model.On this basis,2.5%,5%and 10%Zhenwu decoction-containing serum were added to the low,medium and high dose groups of Zhenwu decoction,and a normal group was set up.The changes of cell activity were assessed by MTT method and LDH method.The contents of TNF-α,IL-6,IL-10,SOD,CAT,GSH-Px,ROS and MDA in each group were detected by ELISA.The apoptosis of each group was detected by flow cytometry.The mRNA and protein expressions of Bax,Bcl-2,caspase-3,caspase-9 and Nrf2/HO-1 pathway were detected by RT-qPCR and Western blot,respectively.Results Compared to the normal group,the model group of HGEC exhibited increased levels of inflammatory cytokines,enhanced oxidative stress response and aggravated apoptosis;after intervention with various doses of Zhenwu decoction,the inflammatory levels in HGEC were reduced,oxidative damage and apoptosis were effectively ameliorated,and the mRNA and protein expression levels of the Nrf2/HO-1 signaling pathway were upregulated.Conclusions Zhenwu decoction can protect HGEC from LPS-induced inflammation and oxidative damage and improve apoptosis.The mechanism may be related to the activation of Nrf2/HO-1 signaling pathway.
