基质分解素-1在胶质瘤组织中的表达研究

摘　　要：目的　了解胶质瘤患者基质分解素 1基因表达的情况。方法　 3 6例脑胶质瘤患者 ,术中取瘤组织及正常组织 (其中Ⅰ～Ⅱ级胶质瘤 18例 ,Ⅲ～Ⅳ级胶质瘤 18例 ,18例正常组织 ) ,提取组织总RNA ,用共扩增逆转录定量PCR方法测定基质分解素 1基因表达水平。结果　Ⅲ～Ⅳ级胶质瘤组织基质分解素 1(MMP3 )的基因表达 (1.0 7± 0 .2 9)明显高于Ⅰ～Ⅱ级胶质瘤 (0 .5 3± 0 .0 86)及正常脑组织 (0 .2 1± 0 .0 2 3 ) ;胶质瘤组织MMP3 基因表达水平与肿瘤大小无关。结论　基质分解素 1在胶质瘤的浸润中可能起重要作用。Objective To invesgtigate stromelysin 1(MMP 3) gene expression in patients with glioma.Methods Glioma tissue and normal brain tissue were obtained from 36 patients with glioma and the total RNA was isolated from the tissues.Stromelysin 1 gene expression level was measured by quantitative analysis of reverse transcription polymerase chain reaction.Results Stromelysin 1 gene expression level in grade Ⅲ～Ⅳ glioma tissue was much ihgher than that of grade Ⅰ～Ⅱ and normal brain tissue.MMP 3 gene expression level had no correlation with tumor size.Conclusion Stromelysin 1 might play an important role in the invasion of malignant glioma.
