卵巢肿瘤中VEGF、VEGFmRNA、TP和TPmRNA的表达及其临床意义

摘　　要：目的:探讨卵巢肿瘤中血管内皮生长因子(VEGF)、VEGFmRNA、胸腺嘧啶脱氧核苷磷酸化酶(TP)和TPmRNA的表达及其临床意义。方法:采用免疫组化法检测卵巢肿瘤VEGF、TP的表达、用原位杂交法检测VEGFmRNA、TPmRNA。结果:35例卵巢癌中,VEGF和VEGFmRNA的阳性率明显高于良性者(74.28%vs42.8%,57.1%vs28.5%,P<0.01)。卵巢浆液性癌和混合性上皮癌的阳性表达率明显高于粘液性癌。随FIGO临床期别的增高,VEGF的阳性率增高(P<0.05)。卵巢癌患者中,腹水>500ml的VEGF阳性率明显高于腹水<500ml者(91.3%vs41.6%,P<0.05),TP和TPmRNA的阳性率明显高于良性者(71.4%vs38.0%,48.5%vs23.4%,P<0.01)。有淋巴结转移者TP表达率明显高于无淋巴结转移者(100%vs61.5%,P<0.05)。结论:VEGF和TP通过自分泌或旁分泌的机制在卵巢癌血管新生中起重要作用,其表达与卵巢癌患者的预后有关。Objectives:To study the expression of VEGF,TP and their mRNA in ovarian tumors as well as their clinical significance.Methods:The expression of VEGF and TP was examined by im-munohistochemical assay while the VEGFmRNA and TPmRNA were measured by ISH in35cases of o-varian cancer and21cases of benign ovarian tumors.Results:The positive expression rates of VEGF and VEGFmRNA were significantly higher in ovarian cancer than in benign tumors(74.28%vs42.8%,57.1%vs28.5%,P<0.01).In cases of serious ovarian cancer and mixed epithelial cancer,the expression rate of VEGF was markedly higher than in those of mucous cancer(P<0.05).There was a close relationship be-tween the expression of VEGF and the FIGO clinical stages of the disease(P<0.05).As far as cancer pa-tients with ascites are concerned,the positive rate of VEGF expression was higher in those with the liquid volume being or above500ml than in those with the liquid less than500ml (91.3%VS 41.6%,P<0.05).The expression rates of TP and TPmRNA in malignant tumor were significantly higher than in the benign ovarian tumor (71.4%vs38.0%,48.5%vs23.4%,P<0.01)The expression rate of TP with lymphonodus metastasis was noticeably higher than in those without lymphonodus metastasis(100%vs61.5%,P<0.05).Conclusions:VEGF and TP play important roles in the angiogenesis of ovarian carcinomas by autocrine or paracrine mechanism.The expression of VEGF and TP is associated with the prognosis of patients with o-varian cancer.
