摘 要:采用正交实验法对蒲黄花粉中多糖的水提取条件进行研究 ,得出最佳提取条件为 :加水比 1 :7,提取温度 60℃ ,提取时间 4h ,提取次数两次 .并对蒲黄花粉进行了酶法破壁条件研究 ,得出最佳破壁条件 :果胶酶用量 30 0u/g花粉 ,纤维素酶用量 1 2 0u/g花粉 ,酶解最适温度 45℃ ,酶解时间 4h .结果表明 ,酶解法与一般水提取法相比 ,提取率由 5 0 .5 0 %提高到 86.5 3%以上 .对提取得到的物质进行了红外光谱分析 ,证明是一种多糖 .In this paper,the method of extraction of polysaccharide from pollen has been studied by orthogonal test. According to the results, we got the best conditions: 1∶7 of proportion of adding water, 60℃ of extraction temperature, 4hr and extraction 2 times. We also studied the method of wallbreaking with enzyme, and found the optimum conditions: pectase 300u/g pollen, cellulase 120u/g pollen, the suitable temperature of hydrolysis is 45℃ and time is 4hr. As a result, the method of extraction by enzyme is better than that with water, the extraction rate increase from 50.50% to 86.53%. The compound was analyzed by IR spectrum, and found it was a polysaccharide.
