摘  要:本研究用MDR_1基因为探针,用Northern blot的方法观察了肝癌组织癌旁肝组织及化疗前后肝癌组织之间MDR_1基因表达的差别。结果发现:肝癌组织中MDR_1基因的表达高于癌旁肝组织;化疗后肝癌组织中MDR_1基因的表达也高于未化疗肝癌组织。提示MDR_1基因的过量表达可能与肝癌固有的和获得性抗药性密切相关。本文还报道了用多聚酶链反应定量分析肝癌组织中MDR_1基因表达的方法。We have cloned full-length MDR1 cDNA from human normal liver tissue in previous study. Using this MDR1 cDNA as probe, we observed the MDR1 gene expression in human hepatocellular carcinoma treated with and without chemotherapy by Northern blot. The results showed that expression of MDR1 gene in hepatocellular carcinoma tissues was higher than that in their adjacent normal liver tissues; and enhanced MDR1 gene expression was also observed in hepatocellular carcinoma treated with chemotherapic agents. We also explored a method for quantitative analysis of MDR1 gene expression in hepatocellular carcinoma by polymerase chain reaction. This study suggests that overexpression of MDR1 gene may be responsible for the intrinsic and acquired drug resistance of human hepatocellular carcinoma, and PCR is a preferable method for quantitative analysis of MDR1 gene expression in hepatocellular carcinoma.
