几种寡糖结构在大肠癌及腺瘤中表达的比较

摘　　要：目的 :为了探讨大肠癌发生过程中寡糖表达模式的变化 ,从而为进一步研究有关糖蛋白分子表达改变找出线索。方法 :采用多种组织学类型的大肠癌 84例 ,大肠腺瘤 5 2例 ,分别进行连续石蜡切片SLex 免疫组化染色和T、ST、Tn以及STn的凝集素组化染色。观察和比较它们在癌与腺瘤细胞浆和细胞膜 (或腺腔缘 )阳性率的差别。结果 :①除了T和Tn胞膜染色在癌组织的阳性率 (分别为 2 6 84[30 1%]和 5 3 84[6 3 1%])比腺瘤者 (分别为 8 5 2 [15 4 %]和 18 5 2 [30 8%]高 (P <0 0 5 ,P <0 0 1) ,各指标在两者的胞膜和胞浆染色阳性率都没有显著差异。②STn染色 ,腺瘤中胞浆 (+)胞膜 (- )模式占 10 5 2 (19 2 %) ,明显比癌组织中 4 84(4 8%)高 (P <0 0 1)。结论 :随着腺瘤细胞向癌细胞的转化 ,尽管胞浆的表达率无差别 ,但胞膜上可能出现更多的T、Tn和STn表达。Objective: To investigate the conversion of the expression pattern of the oligosacchrides during the development of colorectal carcinoma, and find clues for the further research of related glycoproteins. Methods: 84 cases of colorectal carcinomas of various histological types and 52 cases of colorectal adenoma were used. Of them the paraffin embedded serial tissue sections were immunohistochemically stained for SLe x, and affinity histochemically stained with lectins for T, ST, Tn and STn. The positive ratios were then compared of the cytoplasmic and membranous expression of these antigens of the carcinomas and adenomas. Results: ①Eexcept that the ratios of T and Tn expression on cell membrane (lumen border) of carcinomas (26/84 and 53/84, respectively) are higher than those of adenomas (8/52 and 18/52, respectively) (P<0.05, P<0.01, respectively), no significant distinction of the ratios between the carcinomas and adenomas was found. ②For the staining of STn, the expression pattern of cytoplasm (+) membrane (-) of the adenomas bears a ratio of 10/52 (19.2%), significantly higher than that of the carcinomas (4/84)(P<0.01). Conclusion: As adenomous cells transform to cancerous cells, cell membrane may express more T, Tn and STn, even though the cytoplasmic expression shows no distinction.
