摘  要:目的 评价外源性PTEN抑癌基因对人高转移性粘液表皮样癌细胞系M 3SP2药物敏感性的影响。方法 用脂质体介导法将PTEN抑癌基因导入M 3SP2细胞系 ,以空载体转染细胞为对照 ,然后应用MTT比色法测定两种癌细胞对 10种常用抗癌药物敏感性。结果 与对照细胞比较 ,PTEN基因转染细胞对 6种抗癌药物甲氨喋呤、重组人干扰素 -γ、丝裂霉素、多西环素、平阳霉素和氟脲嘧啶的相对抗肿瘤活性呈不同程度地增强 ,药物增敏比增加了 1.5~ 5 2 .1倍。结论 外源性野生型PTEN抑癌基因能增强粘液表皮样癌细胞对常用抗癌药物的敏感性。Objective To evaluate exogenous tumor suppressor gene PTEN on in vitro chemosensitivity of human highly metastatic mucoepidermoid carcinoma cell line M3SP2.Methods Retrovirus expression vector containing wild-type PTEN gene or control vector were introduced into the M3SP2 cells by using cationic liposome, and PTEN transfected cells were named as M3SP2-PTEN, the control vector transfected cells as M3SP2-pBp.The chemosensitivity of the above two cancer cell lines to 10 kinds of anticancer drugs was determined by MTT assay.Results Comparing with the control cells, the PTEN transfected cells revealed more sensitivity to anticancer drugs, such as methotrexate, interferon-γ, mitomycin C, doxycycline, and pinyangmycin, and the sensitizing enhancement ratio increased by 1.5~52.1.Conclusion Introduction of PTEN gene into highly metastatic mucoepidermoid carcinoma cell line can enhance the cells sensitivity to chemotherapeutic agents.
