用荧光光谱与高效液相色谱法研究广西血竭的α-葡萄糖苷酶抑制活性  被引量:2

摘  要:采用冷浸法对广西血竭进行分离,测定各组分的α-葡萄糖苷酶的抑制活性.使用三维荧光指纹技术对其荧光指纹进行研究,发现抑制率与荧光指纹主峰强度的相关系数为0.9413.说明可用荧光指纹参数表征抑制剂的抑制活性.以三维荧光光谱信息为指导找出荧光强度最高处的激发发射波长对,作为高效液相色谱荧光检测器的检测波长,研究各组分色谱峰面积同其相应α-葡萄糖苷酶抑制率的相关关系,发现其中峰1、峰2、峰1+峰2与其抑制率的相关系数分别为0.9414、0.9678和0.9645.说明这两色谱峰所对应的抑制剂与α-葡萄糖苷酶的抑制活性直接相关,为血竭α-葡萄糖苷酶的抑制活性有效成分的筛选,找到了很好的指示方法和检测手段.After Guangxi dragon's blood was separated into several extracts by cold extraction, their inhibitive activity for αglucosidase was measured. Using the analyzing technique of three dimensional fluorescence fingerprint, it has been found that the position and intensity of major fluorescence peaks have strong correlation with inhibitive capability, and the correlation coefficient is 0.9413. Therefore, parameters of the fingerprint can represent inhibitive activity of inhibitors. According to these fluorescence peaks, the wavelength of fluorescence detector of HPLC was determined. By the reversedphase high performance liquid chromatography (RPHPLC), it can be found that peak 1, peak 2 and peak 1 + peak 2 have direct relationship with inhibitive capability of the extracts, and the correlation coefficients are 0.9414,0.9678 and 0.9645 respectively. This method can be used to screen αglucosidase inhibitors and replace enzyme experiments.
