摘 要:本文应用免疫细胞化学技术对新研究成功的结肠癌相关抗原LEA在光镜、电镜下进行了较详细的定位观察研究。LEA是一种可溶性抗原,其表达与癌细胞的分化程度密切相关。应用ABC免疫酶标法发现LEA在癌组织内的分布呈明显的异质性,LEA存在于癌细胞腔缘的质膜上,有明确的极性。在培养细胞,LEA均匀分布在膜表面。免疫电镜观察到LEA多集中分布在微绒毛上,金颗粒标记在细胞外被的糖萼成分上。LEA可能是通过微绒毛结构排出到腺腔中去的。The distribution of LEA, a new colorectal carcinoma-associated antigen, was examined in details by immuno- light and immunoelectron microscopy. LEA is a soluble cancer-associated antigen, its expression is highly in rela-tion to the differentiation of cancer cells. Immunoperoxidase assay found that LEA showed heterogeneous distribu-tion in colorectal cancer tissue and existed restrictly on the luminal plasma membrane of cancer cells. On cultured cell surface, however, LEA did not show this polarized distribution. LEA molecules were seen to loealize much more in microvilli by immunoelectron microscopy, the gold particles were labelled to glycocalyx component in cell coat. It was suggested that LEA might be exported from cells via microvilli.
