^(188)Re-硫化锑纳米胶粒的制备及其被兔淋巴结摄取的实验研究

摘　　要：制备了两种不同粒径的硫化锑纳米胶粒 ,研究了1 88Re标记两种胶粒的条件以及标记物被兔窝淋巴结和腹股沟淋巴结摄取的情况。实验结果表明 ,两种硫化锑纳米胶粒的平均粒径分别为 10 .2nm和 4 4 .7nm。当SnCl2 ·2H2 O的浓度为 4mg mL ,抗坏血酸的浓度为 6mg mL时 ,加入适量的1 88ReO-4淋洗液 ,6 0℃水浴加热 6 0min ,标记率均大于 95 % ,而且有较好的体外稳定性。从正常的新西兰大白兔双足趾蹼皮下分别注射两种标记物 ,γ探针探测的结果表明 ,窝淋巴结对 10 .2nm和 4 4 .7nm胶粒的摄取在注射后 1.5— 2h之间达到最大值 ,分别占各自注入量的17.4 %和 19.4 % ;腹股沟淋巴结对 10 .2nm和 4 4 .7nm胶粒的摄取在注射后 2h左右达到最大值 ,分别占各自注入量的 4 .7%和 4 .2 %。Two different particle sizes of antimony sulfide nanocolloids were prepared and labeled with 188 Re. Their uptake by rabbit' s popliteal lymph node and inguinal lymph node was also studied. Results showed that the mean diameters of the two kinds of particles were 10.2nm and 44.7nm respectively. The labeling condition was optimized. When 4mg/mL SnCl 2·2H 2O, 6g/L Vit C and 1mL nanocolloids were reacted in 60℃ water bath for 60min, the labeling yield were both greater than 95% and excellent in vitro stability was also observed. White New Zealand rabbits were injected subcutaneously on the dorsum of the hind foot with two kinds of 188 Re-Sb 2S 3. The uptake by popliteal lymph node and inguinal lymph node were measured quantitatively by using a gamma detecting probe. Results showed that the highest uptake of 10.2nm and 44.7nm nanocolloids by popliteal lymph node reached 17.4% and 19.4% of the total 1.5 to 2h post injection, respectively; the highest uptake by inguinal lymph node reached 4.7% and 4.2% of the total about 2h post injection, respectively.
