摘 要:目的 观察肝素和氢化可的松对人非小细胞肺癌(NSCLC)移植瘤模型的抗血管生成作用。方法Balb-c裸鼠30只 ,均接种Calu -6细胞 ,随机分为3组 ,每组10只。A组 :第10~30天、第40天至实验结束饮水内加入肝素500IU/ml、氢化可的松0.45mg/ml。B组 :第10~14天、第31~35天腹腔内注射卡铂10mg/(kg·d)。C组 :对照组 ,不进行任何治疗。观察各时点肿瘤体积的变化 ,并检测细胞凋亡、增殖细胞核抗原(PCNA)、血管内皮生长因子(VEGF)蛋白、VEGFmRNA阳性细胞数和血管内皮细胞粘附分子 -1(CD31)染色阳性血管数等。 结果 A组在治疗第2、3、4、8周时肿瘤体积均小于C组(均P<0.05) ,第2周时肿瘤体积大于B组(P<0.05)。A组在凋亡细胞、PCNA、VEGF蛋白与VEGFmRNA阳性细胞数和CD31阳性血管数等方面都与B、C组有明显差别(均P<0.05)。 结论 肝素和氢化可的松联合应用能够有效抑制Calu -6细胞在体内的生长。Objective To observe the anti _ angiogenetic effect of heparin and hydrocortisone in Calu-6 human non _ small _ cell lung cancer(NSCLC) of nude mice.Methods Xenograft tumors were established by subcutaneous injection of Calu _ 6 cells into 30 nude _ mice.The animals were divided randomly into 3 groups:group A,the mice were administrated with heparin (500IU/ml) and hydrocortisone (0.45mg/ml) in the drinking water from the 10th day to the 30th day and from the 40th day to the end of the trial;group B,the mice were treated with carboplatin 10mg/(kg·d) at 10th to 14th and 31st to 35th day;group C,the mice were injected with Calu-6 cells and without any treatment.The volumes of tumors were observed.The number of apoptosis cells and VEGF (vascular endothelial growth factor) -positive cells and VEGF mRNA and proliferating cell nuclear antigen (PCNA) and the number of vascular endothelial cell adhesion molecule-1 (CD 31) positive vascular were detected. Results The mean tumor volume of group A had significant difference with group C at the time of 2nd,3rd,4th and 8th week (P<0.05).The number of apoptosis cells,PCNA,the number of VEGF- positive cells and VEGF mRNA and the positive vessel number of CD 31 of group A had significant difference with those of group B and C (P<0.05).Conclusion The combination of heparin and hydrocortisone can effectively inhibit the growth of xenograft Calu-6 cells in vivo.
