摘 要:目的 探讨白血病细胞株的固有活性氧(ROS)水平与细胞对三氧化二砷(As2O3)促凋亡作用的敏感性之间的关系。方法 用2μmol/L As2O3作用于四种髓系来源的人白血病细胞株(NB4、HL60、K562、U937),证实As2O3促凋亡敏感性在四种细胞之间的差异。然后在不加As2O3情况下,用活性氧捕获剂双氢-乙酰乙酸二氯荧光黄(DCFH-DA)或双氢罗丹明123(DHR)捕获ROS,通过流式细胞仪检测细胞的ROS水平。结果四种白血病细胞凋亡敏感性与ROS水平存在对应关系,由高到低的顺序为:NB4、HL60、K562、U937。结论 白血病细胞固有的ROS水平与细胞对As2O3诱导凋亡的敏感性有关。Objective To explore the association of the inherent cellular level of reactive oxygen species (ROS) with the sensitivity of the leukemia cells to arsenic trioxide ( As2O3). Methods The difference was confirmed among four leukemic cell lines, NB4, HL60, K562, U937 , concerning their sensitivity to apoptosis induction by 2μmol/L of As2O3; 6 - carboxy - 2 ' , 7 '- dichlorodihydrofluorescein diacetate ( DCFH - DA) or dihydrorhodamine 123( DHR) , respectively used as ROS captures, was incubated with the cells in absence of As2O3 administration and ROS was thus measured by flow cytometry. Results The sensitivities of four leukemia cell lines are in accordance with the level of inherent cellular ROS. Conclusion The difference in inherent cellular ROS level of leukemia cells is associated with the cellular sensitivity to apoptosis induction by As2O3.
