摘  要:目的 通过nm2 3 H1的转化及导入BcaCD885细胞株 ,建立稳定、高效、低毒的转染方法 ,观察nm2 3 H1对BcaCD885细胞株侵袭转移能力的影响。方法 ①利用基因转化技术 ,制备高纯度的nm2 3 H1真核表达质粒 ;②利用阳离子脂质体介导的转染技术 ,完成转染方法的建立 ;③利用免疫组化技术 ,检测转染前后的NDPKA的表达 ;④利用transwell小室和冲刷实验 ,观察转染前后细胞的侵袭、粘附、趋化运动能力的变化。结果 ①使用重组的pCMV Neo Bam的真核表达载体 ,将nm2 3 H1转染口腔癌细胞 ,并获得稳定表达 ;②发现BcaCD885细胞株nm2 3 H1基因的转染前后表达水平有明显差异 ;③转染后的BcaCD885细胞侵袭、粘附、趋化运动能力均显著降低。结论 nm2 3Objective To transfect nm23 H1 into the BcaCD885 cell lines in order to get safe high efficiency and low toxicity, and to find out whether nm23 H1 could affect the invasion and metastases ability of BcaCD885 cell lines. Methods Lipofect was used to transfect nm23 H1 into BcaCD885 cell lines;immuhistochemistry was used to dectect the difference expression of nm23 H1 between transfected and non transfected cell lines; then transwell room and wash way were used to detect the difference of invision and metastases ability between transfected and non transfected cell lines. Results PCMV NEO BAM system gave the stability expression of nm23 H1;there was significant differenct NDPKA expression between transfected and non transfected BcaCD885 cell lines; the invasion and metastases ability of transfected BcaCD885 cell lines decreased obviously. Conclusion nm23 H1 can inhibit the metastases of BcaCD885 cell lines significantly.
