摘  要:【目的】 观察重复过敏原吸入刺激致敏的大鼠气道细胞DNA合成和气道壁结构的改变 ,并探讨气道重塑反应发生的机制。 【方法】 应用SD大鼠建立哮喘模型 ,采用双标免疫组化技术和计算机图像分析系统对气道细胞的DNA合成和气道重塑进行研究。 【结果】 ①模型组气道平滑肌细胞和上皮细胞的DNA合成Brdu阳性计数明显高于正常对照组 (P <0 .0 1,P <0 .0 5 ) ;②模型组气道平滑肌细胞和上皮细胞DNA合成Brdu阳性计数和气道直径呈高度正相关 (P <0 .0 1,P <0 .0 5 ) ,正常对照组无明显相关性 (P >0 .0 1,P >0 .0 5 ) ;③模型组上皮层厚度明显大于正常对照组 (P <0 .0 1) ,平滑肌层厚度、气道直径和α 平滑肌肌动蛋白阳性区面积和正常对照组差异无显著性 (P>0 .0 5 )。 【结论】 变态反应性炎症所致的微环境改变可以导致气道细胞DNA合成增加以及细胞增生 。To study DNA synthesis of airway cells after being stimulated with allergen and investigate airway remodelling. Double staining immunohistochemical techniques was used to determine DNA synthesis of airway cells.Brdu incorporation into airway smooth muscle(ASM) and epithelium was quantified employing computer asisted image analysis. ①The Brdu indices in ASM and epithelium of moldel group were both higher than those of the control group( P <0.01, P <0.05).②In model group,there was positive linear correlation of the Brdu indices in the ASM and epithelium to airway diameter( r=0.782 8,P <0.01; r=0.585 2,P <0.05),no correlation was found between Brdu indices and airway size in control group( r= 0.377 58,P >0.01; r= 0.520 8,P >0.05).③The epithelial thickness of the model group was significantly higher than that of control group( P <0.01).There was no significant difference in airway diameter,thickness of ASM and area of positive with α smooth muscle action between two groups( P >0.05). [Conclusions] DNA synthesis in ASM and epithelial cells were increased in sensitized SD rats following repeated allergen challenges,suggesting an increase cell proliferative response to allergen,which might lead to airway remodelling.
