摘 要:目的观察顺铂对豚鼠耳蜗毛细胞的损害 ,并阐述一氧化氮 (nitricoxide ,NO)在其耳毒性机制中的作用。方法将 2 0只豚鼠随机分成两组 ,试验组腹腔注射顺铂 0 .5ml,对照组给予同等量的生理盐水 ,5天后做听性脑干反应 (auditorybrainstemresponse ,ABR)测听 ,此后处死动物 ,取出耳蜗 ,应用免疫组化技术、光镜及扫描电镜观察耳蜗毛细胞的损害。结果试验组动物测试ABR听阈明显提高 ,光镜下见耳蜗毛细胞明显变性 ,诱导型一氧化氮合酶阳性染色 ,电镜下见毛细胞的纤毛散乱、倒伏甚至散失。结论顺铂对豚鼠耳蜗毛细胞有明确的损害作用 ,而NO在其耳毒性机制中发挥着重要作用。ObjectiveTo observe impairment of cochle ar hair cell in guinea pig after using Cisplatin and study the mechanism of nitric oxide(NO) effect on Cisplatin oto toxicity.MethodsTwenty guinea pigs were divided randomly into two groups: control group and experimental group. 0.5 ml Cisplatin were injected to abdominal cavity in experimental guinea pigs, 0.5 ml physiological saline(N.S) were injected to abdominal cavity in control animals.Five days later,all guinea pigs were killed. The cochlea of guinea pig were removed and cochlear hair cell were observed by means of light microscope and scanning electron microscope (SEM). ResultsThe hearing threshold of ABR examination were increase in experimental group.Light microscope showed that there were denatured cochlear hair cell and inducible nitric oxide synthase positive stain.Scanning eletron microscope showed that the cilia of the majority cochlear hair cell were disordered,fallen,even disappeared.ConclusionCochlear hair cell are impaired definitely after using Cisplatin and NO play an important role in Cisplatin oto toxicity mechanism.
