三氧化二砷诱导Cloundman S91恶性黑素瘤细胞凋亡的研究

摘　　要：目的:探讨三氧化二砷(As_2O_3)对小鼠黑素瘤CloundmanS91细胞系的作用。方法:四氮唑(MTT)还原法检测As2O3对S91黑素瘤细胞存活率的影响,通过形态观察、流式细胞仪分析、DNA凝胶电泳研究As2O3诱导S91细胞凋亡的情况。结果:S91细胞经As2O3处理后,存活率降低,并与药物浓度呈正相关;荧光显微镜下可见明显的凋亡细胞;细胞周期G1期前出现凋亡峰;DNA凝胶电泳显示有规律的梯状条带。结论:3～5μmol/L的As2O3诱导S91黑素瘤细胞凋亡。Objective:To investigate the biological effect of arsenic trioxide(As 2 O 3 )on mice melanoma Cloundman S91cells.Meth ods :MTT re duction assay,morphologic change fo the cells,flow cytometry analysis technique,agarose gel electrophoresis were performed.Re sults:The inhibition of As 2 O 3 on the growth of S91cell line was concentration and time-dependent.The cells treated with3-5μmol/L As 2 O 3 for72hours showed a typical apoptotic morphology and hypodiploid peak before G 1 phase.DNA of S91cells treated by4-5μmol/L As 2 O 3 for48hours appeared a ladder pattern characteristic of apoptosis.Conclu sions:As 2 O 3 can in duce melanoma Cloundman S91cell apoptosis at the concentration of3-5μmol/L.
